Summary: A nickel-catalyzed stereoselective asymmetric intramolecular reductive coupling of N-1,6-alkynones was reported, achieving efficient synthesis of versatile functionalized chiral pyrrolidines with high yields, excellent stereoselectivity, and enantioselectivity. The developed reaction is applicable to a broad substrate scope and can be scaled up to gram scale without loss of enantioselectivity. The study also investigated ligand effects and reaction mechanism, suggesting wider applications in organic synthesis and chemical biology, and potential for further explorations in new research fields.

Summary: This paper presents an efficient synthesis method for substituted (Z)-N-allyl sulfonamides via a three-component tandem reaction catalyzed by Pd(PPh3)4. The optimal catalyst, base, and solvent used were 2.5 mol % Pd(PPh3)4, K2CO3, and THF, respectively. The desired products were obtained in an overall yield of 30-83%. Mechanistic investigations showed that the control of the formation of the single (Z)-isomer relied on the formation of a six-membered palladacycle intermediate.

Summary: An efficient Ru doped hydrotalcite catalyst was used for the N-alkylation of benzamides and sulfonamides with alcohols via borrowing hydrogen catalysis. The reaction showed good to excellent yields with various primary alcohols. Mechanistic studies revealed that the reaction proceeds via a borrowing hydrogen pathway. The catalyst also demonstrated the capability to directly synthesize quinazolinone and exhibited successful recyclability and high reactivity.

Summary: In this study, an electron-deficient alkene embedded with an oxidizing function/leaving group was used as a rare and nontraditional C-1 synthon to achieve a redox-neutral Rh(iii)-catalyzed chemo- and regioselective [4 + 1] annulation of benzamides for the synthesis of functionalized isoindolinones. This method demonstrated a broad substrate scope, good to excellent yields, excellent chemo- and regioselectivity, good tolerance of functional groups, and mild external-oxidant-free conditions.

Summary: An enantioselective Ni-catalyzed syn-hydrocyclization reaction was developed for the synthesis of α-hydroxy γ-lactams. By using Ni(OTs)(2)·6H(2)O/(S,S)-Me-Duphos as a precatalyst and (EtO)2MeSiH as a hydride source, a wide range of enantioenriched γ-lactams with fully substituted stereogenic centers were obtained in 32-84% yields with 87.5:12.5-97:3 er. This research provides a regioselective functionalization of alkynes and offers an efficient strategy for accessing functional group-enriched chiral heterocycles.

Summary: The synthesis of isoindolinones from N-methoxy benzamides and saturated ketones has been achieved through a bimetallic tandem catalytic annulation. The reaction is catalyzed by a Rh/Cu-cocatalytic system, involving Cu-catalyzed dehydrogenation of ketones and Rh-catalyzed direct C-H functionalization with the assistance of the N-methoxy amide group as an oxidant to regenerate the Rh catalyst. This method is compatible with a wide range of substrates and functional groups, providing an alternative strategy for obtaining diverse isoindolinones.

Summary: An iron(II)-catalyzed bisphosphorylation cascade cycloisomerization of readily accessible gamma-hydroxy ynones and diphenylphosphine oxides is reported. This strategy provides a variety of valuable polysubstituted bisphosphorylated dihydrofuran scaffolds via the construction of two C-P bonds and one C-O bond within a single procedure. The developed reaction system demonstrates good functional group compatibilities with considerably low catalyst consumption, and can be further scaled up to gram quantities in satisfactory yields.

Summary: This study presents the design and synthesis of sulfated disaccharide inhibitors based on IdoA(2S)-GlcNS(6S), which showed potent inhibition of Sulf-2. The results suggest that IdoA(2S)-GlcNS(6S) is the shortest fragment size required for effective inhibition of Sulfs, and a trisulfated disaccharide is identified as the minimal fragment size of heparan sulfate for effective endosulfatase inhibition.

Summary: The self-adaptability of galvanostatic electrolysis was found to be helpful in a multistage chemo- and diastereoselective electrochemically promoted cyclodimerization of chalcones. Through a series of reductive events, densely functionalized cyclopentanes with five contiguous stereocenters were obtained (25 examples, yields up to 95%, dr values up to >20:1). Further experimental and electrochemical investigations indicated the crucial role of dynamic kinetic resolution of the aldol intermediate in the reaction mechanism.
