Summary: This study aimed to investigate the core active constituents and pharmacological mechanism of Pinelliae Rhizoma Praeparatum (PRP) against cold asthma (CA). The results showed that PRP suppressed the allergic airway inflammation of CA by regulating the balance of Th1 and Th2 cytokines and possibly involving the PKC/EGFR/MAPK/PI3KAkt signaling pathway.

Summary: The aim of this study is to investigate the active compounds and molecular mechanism of traditional Chinese medicine Yangqing Chenfei formula (YCF) in treating silicosis. The results showed that YCF can improve lung function and pathological changes in silicotic rats, inhibit fibroblast activation, and suppress the TGF-β/Smad3 signaling pathway. YCF6, the effective fraction derived from YCF, can significantly inhibit fibroblast activation induced by TGF-β.

Summary: This study developed a transcriptomic signature to identify KEAP1/NFE2L2-activated NSCLC by accurately detecting KEAP1/NFE2L2 mutational cases and discovering alternative genomic activators. The signature showed good performance in predicting patient survival and has the potential to be used for selecting patients with a constitutively active KEAP1/NFE2L2 pathway.

Summary: The study showed that osimertinib could effectively suppress the proliferation of a subset of EGFR-negative GBM cells by inhibiting MNK kinase activities and eIF4E phosphorylation. Additionally, oral administration of osimertinib significantly suppressed tumor growth and could be a potential new drug candidate for EGFR-negative GBM patients.

Summary: In this study, the effective components and underlying mechanisms of Quince (Cydonia oblonga Mill, COM) extract against atherosclerosis were identified. Using UHPLC-Q-TOF-MS/MS, 14 serum components of COM extract were identified. Network pharmacology analysis predicted 573 targets, including 224 atherosclerosis-specific targets. The key targets included GSK3 beta, ESR1, EGFR, and HSP90AA1, and the key signaling pathway was the PI3K-Akt signaling pathway. COM extract exhibited hypolipidemic, anti-oxidative, anti-inflammatory, anti-thrombotic, and vascular endothelium protective effects in a rat model of atherosclerosis. These effects may be related to the inhibition of the EGFR/PI3K/AKT/GSK-3 beta signaling pathway.

Summary: This study explores the potential anti-HCC mechanism of Yupingfeng San (YPFS) in liver cancer treatment through systems pharmacology and high throughput sequencing-based high throughput screening (HTS2) technology. The results show that YPFS possesses anti-proliferative and pro-apoptotic activities in treating HCC by inhibiting the MAPK signaling pathway. P38 is one of the critical targets of YPFS in treating HCC, which may be directly bound and inhibited by 4'-O-glucosyl-5-O-methylvisamminol, a compound derived from YPFS.

Summary: Six novel circRNAs were identified and validated in ameloblastoma. The interaction between hsa_circ_0089153/hsa-miR-608 and downstream genes was confirmed. In vitro experiments demonstrated the suppressive function of miR-608 on EGFR and p53. Functions were mainly enriched in MAPK signaling pathways regulating AB progression.

Summary: The study reveals that up-regulation of PTPN21 is correlated with glioma and predicts poor survival rate in patients. Silencing PTPN21 has significant anti-cancer effects in glioma cells, inhibiting proliferation, arresting cell cycle, suppressing metastasis, and enhancing chemosensitivity. Mechanistically, PTPN21 mediates the EGFR/PI3K/AKT pathway in glioma.

Summary: This study revealed that exosomal DNAJB11 promotes cell development in pancreatic cancer cells by regulating the expression of HSPA5 and activating the endoplasmic reticulum stress pathway. Additionally, exosomal DNAJB11 can regulate the expression of EGFR and activate the MAPK signaling pathway. Clinical studies suggest that exosomal DNAJB11 has the potential to serve as a diagnostic biomarker and therapeutic target for pancreatic cancer.

Summary: Gliomas are debilitating malignant brain tumors with limited response to therapies, driven by molecular abnormalities like mutations in regulatory networks. The MAPK pathway, particularly p38 MAPK activation, plays a crucial role in glioma cell invasion and metastasis, and is associated with tumor grade and chemotherapy resistance, underlining its potential as a therapeutic target.
