Summary: Bone marrow stem cells (BMSCs) are potential seed cells for bone tissue engineering, but cell senescence could affect their therapeutic effects. This study aimed to explore the transcriptomic differences between uncultured and passaged BMSCs and identify a practical anti-aging target gene. Through flow cytometry analysis, PDGFR-alpha(+)SCA-1(+)CD45(-)TER119(-) cells were sorted as BMSCs. Various cellular senescence phenotypes and associated transcriptional alterations during different cell culture processes were studied. Imprinted zinc-finger gene 1 (Zim1) was found to play a critical role in anti-aging, and its combination with Gelatin methacryloyl (GelMA) reduced senescence markers and increased telomerase activity. The combined application of Zim1 and hydrogel showed potential for inhibiting the senescence of BMSCs during in vitro expansion.

Summary: Periodontal tissue regeneration can be achieved by transplanting clumps of mesenchymal stem cells (MSCs) and extracellular matrix (ECM) complexes (C-MSCs) without artificial scaffold. In a rat model, C-MSCs transplantation successfully induced periodontal tissue regeneration, with the donor cells directly involved in tissue formation. This study demonstrates a promising scaffold-free MSCs transplantation strategy for periodontal disease.

Summary: This study investigated the relationship between masticatory disorders and diabetic microvascular complications. The results showed that patients with type 2 diabetes who had diabetic neuropathy had significantly reduced masticatory efficiency. Effective mastication is an important factor in successful diet therapy for diabetes, and for patients with diabetic neuropathy, it may be necessary to combine individualized therapies from dentists and nutritionists, taking into consideration the level of masticatory dysfunction, to prevent the progression of diabetic complications.
