Targeted drug delivery systems: synthesis andin vitrobioactivity and apoptosis studies of gemcitabine-carbon dot conjugates

Gemcitabine (GEM) is used to treat various cancers such as breast, pancreatic, non-small lung, ovarian, bladder, and cervical cancers. GEM, however, has the problem of non-selectivity. Water-soluble, fluorescent, and mono-dispersed carbon dots (CDs) were fabricated by ultrasonication of sucrose. The CDs were further conjugated with GEM through amide linkage. The physical and morphological properties of these carbon dot-gemcitabine (CD-GEM) conjugates were determined using different analytical techniques.In vitrocytotoxicity and apoptosis studies of CD-GEM conjugates were evaluated by various bioactivity assays on human cell lines, MCF-7 (human breast adenocarcinoma), and HeLa (cervical cancer) cell lines. The results of kinetic studies have shown a maximum drug loading efficacy of 17.0 mg of GEM per 50.0 mg of CDs. The CDs were found biocompatible, and the CD-GEM conjugates exhibited excellent bioactivity and exerted potent cytotoxicity against tumor cells with an IC(50)value of 19.50 mu g ml(-1)in HeLa cells, which is lower than the IC(50)value of pure GEM (similar to 20.10 mu g ml(-1)).In vitrostudies on CD-GEM conjugates demonstrated the potential to replace the conventional administration of GEM. CD-GEM conjugates are more stable, have a higher aqueous solubility, and are more cytotoxic as compared to GEM alone. The CD-GEM conjugates show reduced side effects in the normal cells along with excellent cellular uptake. Hence, CD-GEM conjugates are more selective toward cancerous cell lines as compared to non-cancerous cells. Also, the CD-GEM conjugates successfully induced early and late apoptosis in cancer cell lines and might be effective and safe to use forin vivoapplications.

Structural and functional insight into thiazolidinone derivatives as novel candidates for anticancer drug design: in vitro biological and in-silico strategies

Summary: The synthesis of compounds with a thiazolidinone pharmacophore was achieved, and their anticancer activities were evaluated. Compounds 2c and 2d exhibited significant cytotoxicity against MCF-7 and HeLa cell lines by inducing apoptosis. Molecular docking and dynamic simulations provided further support to these findings, and computational studies suggested the potential of these compounds as lead molecules for the development of novel drug candidates for specific cancers.

Optimization of C50 Carotenoids Production by Open Fermentation of Halorubrum sp. HRM-150

Summary: This study aims to find more cost-effective and manageable ways of producing C-50 carotenoids. The results showed that using natural brine medium and adding glucose, sucrose, and lactose can enhance both biomass and carotenoid productivity. The best conditions for open fermentation were salinity 20-25%, rotation speed 150-200 rpm, and pH 7.0-8.2. The open fermentation of Halorubrum is a promising method for producing C-50 carotenoids.

Assessment of mercury contamination and food composition in commercially important marine fishes in the southern South China Sea

Summary: This study investigated mercury contamination in common commercial marine fish from the South China Sea. The muscle tissue mercury levels of four species were analyzed, and their trophic level was assessed. The results revealed variations in mercury concentrations among species, with Epinephelus heniochus having the highest concentration and Scomberomorus guttatus having the lowest. Factors such as size, food composition, and habitat were identified as the major influences on mercury levels in these fishes.

Characterization and genome analysis of Vibrio phage vB_VhaP_PG11, representing a new viral genus

Summary: A novel marine phage vB_VhaP_PG11 infecting Vibrio hangzhouensis was isolated from offshore waters of Qingdao, China. It is a double-stranded DNA phage and is related to two Vibrio phages with low homology. Phylogenetic analysis revealed that vB_VhaP_PG11 belongs to a new genus named Qingschitovirus in Schitoviridae. This study provides important information for understanding the interactions between Vibrio phages and their hosts and analyzing unknown viral sequences in metagenomic databases.

The Efficacy of Metformin and Exenatide in Polycystic Ovary Syndrome (PCOS) Patients

Summary: Polycystic Ovary Syndrome (PCOS) is a hormonal disorder that affects 5-10% of women in their reproductive age. This meta-analysis evaluates the efficacy of metformin and exenatide in reducing BMI and testosterone levels in PCOS patients. The results indicate that exenatide is effective in reducing BMI and testosterone levels in PCOS patients compared to metformin, but it has no effect on LDL-C and HDL-C levels.

Synthesis and Antioxidant Studies of 2,4-Dioxothiazolidine-5-acetic Acid Based Organic Salts: SC-XRD and DFT Approach

Summary: In this study, two organic salts (1 and 2) were synthesized and characterized by FTIR, UV spectroscopy, and X-ray crystallographic studies. The stability and charge transfer phenomenon of the compounds were confirmed through analysis of natural bond orbitals (NBOs), natural population, frontier molecular orbitals (FMOs), and global reactivity parameters. Molecular docking predicted that the coformer exhibited DNA intercalation reinforced by its salt formation. Antioxidant studies showed that compound 3 has a higher IC50 value than 2,4-D and the highest value among the compounds studied, while compound 2 showed the lowest value.

The development of chiral metal-organic frameworks for enantioseparation of racemates

Summary: In this study, MIL-101 (Cr), an easily activated metal-organic framework with high specific surface area, big pore size, and good stability, was modified by introducing chiral ligands (l-proline, l-thioproline, and l-tyrosine) to develop chiral metal-organic frameworks (CMOFs). The prepared CMOFs showed enantioselectivity towards various racemates, as confirmed by chiral HPLC and polarimetry.

Iron Deficiency Amongst University Graduates; Prevalence, Diagnosis, and Iron Consumption

Summary: Anemia is a global epidemic caused by iron deficiency. This study used anthropometric measurements and a questionnaire to investigate the correlation between iron deficiency, diet, and anemia. The results showed that both underweight and obesity were prevalent among men, and 10% of men with normal blood iron levels had mild iron deficiency. In women, underweight was more common than obesity. The study also highlighted the potential risks and benefits of iron supplements. Understanding the relationship between iron deficiency anemia and specific nutrients is important for maintaining proper nutrient levels and promoting public health.

Genomic diversity and ecological distribution of marine Pseudoalteromonas phages

Summary: This study systematically analyzed the genomes, taxonomic features, and distribution patterns of Pseudoalteromonas phages. A total of 143 Pseudoalteromonas-associated phage genomes were identified and assigned to 47 viral clusters. The prevalence of integrated proviruses and filamentous phages suggest the viral lysogenic life cycle in Pseudoalteromonas. These phages play a role in marine biogeochemical cycles and their abundance and distribution vary across polar areas and the deep sea. This study expands our understanding of marine Pseudoalteromonas phages and their interactions with their hosts.
