Summary: Targeting cancer hallmarks is crucial for improving anti-cancer treatment, but cross-talk between signaling pathways often leads to resistance. This article provides an overview of the molecular interactions between the p53 and Wnt pathways in cancer, including complex feedback loops and reciprocal transactivation, as well as the mutational landscape of genes associated with these pathways.

Summary: Src, a non-receptor tyrosine kinase, plays a critical role in cancer progression for various tissues. It regulates Wnt signaling through multiple signaling molecules, and aberrant regulation of this interaction can lead to the development of various diseases, particularly cancer.

Summary: In this study, we investigated the subcellular localization of EGFR and its mutants in normal and cancer cells, as well as the impact of subcellular localization on its activities using various experimental approaches. Our results suggest that EGFR plays a role in modulating TCF-dependent beta-catenin transcriptional activity in HepG2 cells, and that cytoplasmic and nuclear functions of EGFR are two independent activities.

Summary: EGFR and E-cadherin are major regulators of proliferation and differentiation in epithelial cells, and defects in either can lead to malignancies. The reciprocal interactions between EGFR and E-cadherin contribute to tumorigenesis and cancer cell invasion through modulation of adhesion and activity regulation. Understanding the crosstalk between EGFR and E-cadherin is important for tissue morphogenesis, homeostasis, and cancer progression.

Summary: The study showed that the silencing of SERPINB2 can promote osteogenic differentiation of BMSCs through the Wnt/β-catenin signaling pathway. In vivo experiments demonstrated that silencing SERPINB2 effectively improves bone fracture healing, suggesting that SERPINB2 may be a novel target for bone fracture healing.

Summary: This study discovered that fibrous dysplasia (FD) patient-derived bone marrow stromal cells (FD BMSCs) exhibited significantly higher levels of PTHrP during osteogenic differentiation and had increased proliferation capacity but impaired osteogenic ability. Through the PTHrP/cAMP/PKA axis, PTHrP partially influenced the proliferation and osteogenic capacity of FD BMSCs by overactivating the Wnt/β-Catenin signaling pathway. This study provides new insights into the pathogenesis of FD and its molecular signaling pathways, offering theoretical evidence for potential therapeutic targets for FD.

Summary: Ongoing research has shown that the interaction between macrophages and mesenchymal stem cells (MSCs) is significant. Lysine-specific demethylase 6B (KDM6B) has been found to control the osteogenic differentiation of MSCs. However, the role of KDM6B in bone marrow-derived macrophages (BMDMs) is still controversial. This study demonstrates that KDM6B serves as a positive regulator for osteogenic differentiation by decreasing the M1/M2 ratio both in vitro and in vivo.
