A feedback loop involving MdMYB108L and MdHY5 controls apple cold tolerance

The MYB transcription factors are important for many aspects of plant stress responses. In this study, we isolated and identified an apple MYB gene, MdMYB108L, whose expression is induced by light and cold stresses. An analysis of MdMYB108L-overexpressing transgenic apple calli revealed that MdMYB108L enhances cold tolerance in apple by upregulating MdCBF3 expression. Interestingly, the expression of MdHY5, which encodes an integrator of light and cold signals, was significantly downregulated in transgenic calli. Yeast one-hybrid and electrophoretic mobility shift assays indicated that MdMYB108L positively regulates cold tolerance by binding to the MdCBF3 promoter. Additionally, MdHY5 functions upstream of MdMYB108L, and the resulting increase in MdMYB108L abundance downregulates MdHY5 transcription. The results of this study elucidate a new pathway for the regulation of apple cold tolerance via a feedback mechanism involving MdMYB108L and MdHY5. (C) 2019 Published by Elsevier Inc.

Seismic performance of joint between L-shaped CFST column and H-section beam with lower plate and extended flange plate

Summary: In this study, a new bolted joint configuration was proposed and tested. The results showed that the slippages between the extended flange plate and the beam flange improved the ductility and energy dissipation capacity of the connection. Increasing bolt number on the beam flange significantly increased the joint resistance and stiffness. Additionally, the beam vertical stiffeners limited the development of plastic deformation and reduced local buckling of the beam section. Based on the findings, simplified calculation formulas and design procedures were proposed.

Chemical fumigants control apple replant disease: Microbial community structure-mediated inhibition of Fusarium and degradation of phenolic acids

Summary: This study investigated the effects of different chemical fumigants on Fusarium and phenolic acids in apple replant soil. The results showed that the chemical fumigants disrupted the soil microbial community to varying degrees, and the content of Fusarium and the number of soil microbial taxa were significantly affected. Greater disturbance resulted in a slower recovery rate of the soil microbial community, promoting the transformation of the soil into a disease-suppressing state.

Genome Sequence Resource of Fusarium proliferatum f. sp. malus domestica MR5, the Causative Agent of Apple Replant Disease

Summary: Apple replant disease (ARD) caused by Fusarium proliferatum f. sp. malus domestica (Fpmd) MR5 is a fungal pathogen that leads to annual losses in apple production in China. In this study, the whole-genome sequence of the highly virulent Fpmd MR5 was obtained, revealing potential pathogenicity-related genes and specific virulence factor genes. These findings will contribute to further research on host-pathogen interactions and the development of disease management strategies.

MdMYB305-MdbHLH33-MdMYB10 regulates sugar and anthocyanin balance in red-fleshed apple fruits

Summary: The R2R3-MYB transcription factor MdMYB305 is positively correlated with fruit sugar content but negatively correlated with anthocyanin content in red-fleshed apples. It promotes sugar accumulation and inhibits anthocyanin synthesis through binding with sugar-related and anthocyanin-related genes. MdMYB305 competes with MdMYB10 for the MdbHLH33 binding site to balance sugar and anthocyanin accumulation in fruits.

Analysis of Soil Fungal Community in Aged Apple Orchards in Luochuan County, Shaanxi Province

Summary: The Luochuan area is vital for apple production in China, but the occurrence of apple replant disease (ARD) in aged apple orchards has hindered sustainable development. The study examined the soil fungal community in 14 aged apple orchards in Luochuan and found significant differences in fungal diversity. Harmful fungi, such as Gibberella, Fusarium, and Cryptococcus, were present but varied in abundance across different aged apple orchards. Pathogenic fungi were identified, increasing disease risk and affecting fruit tree growth and development. The unbalanced microbial community structure can be addressed during orchard renewal to mitigate the impact of ARD on the apple industry.

Abscisic acid and regulation of the sugar transporter gene MdSWEET9b promote apple sugar accumulation

The results showed that MdSWEET9b promotes sugar accumulation by transporting sucrose, and MdWRKY9 and MdbZIP23/46 regulate its expression in the ABA signaling pathway, thereby influencing sugar accumulation.

Effect of Hydrogen Peroxide on the Soil Microbial Community Structure and Growth of Malus hupehensis Rehd. Seedlings under Replant Conditions

Summary: Apple replant disease (ARD) is a common problem in apple production, affecting the growth and development of apples. This study investigated the use of hydrogen peroxide to treat replanted soil and its effects on seedling growth and soil microbiology. The results showed that hydrogen peroxide treatment improved seedling growth and reduced the population of Fusarium, while increasing the abundance of Bacillus, Mortierella, and Guehomyces. The best results were obtained with replanted soil + 4.5% hydrogen peroxide. Therefore, hydrogen peroxide can effectively prevent and control ARD.

Analysis of the Postharvest Storage Characteristics of Two New Pear Cultivars 'Shannongsu' and 'Xincixiang'

Summary: 'Shannongsu' and 'Xincixiang' are two new late-ripening pear cultivars with high quality bred by our team. The postharvest storage characteristics of mature pears were studied by storing them at room temperature. Various fruit characteristics were evaluated over time, including firmness, ethylene release rate, aroma component content, softening-related enzyme activity, and gene expression. Both 'Shannongsu' and 'Xincixiang' exhibited excellent storage characteristics, remaining crisp and juicy after 60 days at room temperature, which may be attributed to low levels of ethylene production, aroma synthesis, softening-related activities, and gene expression. This research provides scientific guidance for fruit production and marketing by clarifying the storage characteristics of 'Shannongsu' and 'Xincixiang' at room temperature for the first time.

A nano-liposomal carrier containing p-coumaric acid for induction of targeted apoptosis on melanoma cells and kinetic modeling

TP53INP2 modulates the malignant progression of colorectal cancer by reducing the inactive form of β-catenin

Summary: This study found that low expression of TP53INP2 is associated with poor survival in colorectal cancer (CRC) patients. As the malignancy of CRC progresses, TP53INP2 expression gradually decreases. Knockdown of TP53INP2 promotes CRC cell proliferation and tumor growth. Mechanistically, TP53INP2 deficiency decreases phosphorylation of beta-catenin, leading to increased accumulation and enhanced nuclear translocation and transcriptional activity. Additionally, TP53INP2 sequesters TIM50, inhibiting its activation of beta-catenin. In conclusion, downregulation of TP53INP2 promotes CRC progression by activating beta-catenin.

The four subunits of rabbit skeletal muscle lactate dehydrogenase do not exert their catalytic action additively

Summary: Oligomeric enzymes are known for their higher catalytic rates compared to monomeric enzymes, but the extent of additivity in their activity is still not well understood. This study used tetrameric rabbit lactate dehydrogenase as a model to examine the kinetics of its catalytic action. Surprisingly, when the concentration of the limiting reactant exceeded that of a single subunit, there was a significant slowdown in the enzyme's conformational rearrangements.

The results showed that indium curcumin and vanadyl curcumin exhibited higher binding affinities and stronger inhibitory effects on amyloid fibrillation compared to gallium curcumin.
