Summary: This study aimed to prepare Span 60-based elastic nanovesicles using the adapted ethanol injection method, and evaluate their ability to improve transdermal permeation of tacrolimus. Different concentrations and types of penetration enhancers were assessed for their impact on the physicochemical properties of the nanovesicles. The results showed significant enhancement in in vitro release compared to pure drug suspension.

Summary: This research aims to develop and optimize Raloxifene HCL loaded spanlastics. A nanocarrier was prepared using ethanol injection method and optimized using Box Behnken design. The effects of independent variables on vesicle size, polydispersity index, and encapsulation efficiency were evaluated. The optimized spanlastics formulation showed suitable particle size for skin permeation. Various characterization and release studies were performed. The result indicated that Raloxifene HCL loaded spanlastics could be a better approach for delivering drugs in deeper layers of the skin.

Summary: This study aimed to encapsulate the antifungal drug clotrimazole (CLT) into spanlastics (SPs) to provide controlled ocular delivery. The optimal formulation S1 showed high encapsulation efficiency, elastic vesicles, sustained release profile, and superior inhibition of Candida albicans development compared to CLT suspension. In vivo histopathological examination confirmed the safety of S1 for ophthalmic application in rabbits, demonstrating the efficacy of SPs for ocular delivery of CLT.

Summary: The objective of this research was to encapsulate niflumic acid into bilosomes for transdermal delivery and effective treatment of osteoarthritis. The optimal formulation (B5) contained 5 mg of sodium taurocholate and 5 mg of Brij-93, showing high entrapment efficiency and suitable particle size. B5 gel exhibited higher drug permeation across rat skin compared to NA gel and demonstrated superior efficacy in vivo.

Summary: This study aimed to enhance drug absorption and bioavailability of paroxetine by preparing novel paroxetine-loaded solid lipid nanoparticles (SLNs) based sustained-release transdermal patches. The SLNs showed a particle size range of 113-230 nm and an entrapment efficiency of 85.14%. Paroxetine was evenly distributed in an amorphous form in SLNs, as demonstrated by DSC and PXRD analysis. SLNs formulated patches showed higher drug permeation through the skin and sustained release behavior. Draize patch test revealed no sign of erythema after applying paroxetine-loaded SLN patches.

Summary: In vitro permeation test (IVPT) is commonly used for evaluating topical preparations and transdermal drug delivery systems. However, the storage of ex vivo skin for IVPT is challenging. This study investigated the efficacy of two cryopreservation media, 10% DMSO and 10% GLY, for preserving rat and pig skin at -20°C and -80°C, and found that -80°C storage in 10% DMSO or 10% GLY was optimal for maintaining skin viability and permeability. The results also suggested that skin viability could serve as a potential indicator for IVPT skin.

Summary: This study aimed to enhance the delivery of vanillic acid (VA) for the treatment of ocular inflammation using novel surfactant-based elastic vesicular nanocarriers called Spanlastics. The formulation composed of Span 60 and Tween 80 displayed the smallest particle size and good physical stability. In vivo experiments showed that the VA-loaded Spanlastic formulation significantly reduced inflammation compared to the drug suspension.

Through experimentation and optimization, an O-GL hydrogel with desirable characteristics was obtained, which demonstrated excellent ex-vivo deposition and in-vivo penetration into deep dermal skin layers, making it a promising option for the effective treatment of fungal infections.

This study successfully improved the dissolution rate and bioavailability of FXS using a patient-wise convenient formula.
