Summary: The study aimed to evaluate PTEN regulation and identify targets for relieving chemoresistance in bladder cancer. The expression of YTHDC1 was found to be associated with cisplatin sensitivity in patients with bladder cancer. Decreased YTHDC1 expression promotes cisplatin resistance, while overexpression of YTHDC1 enhances cisplatin sensitivity. Furthermore, reducing YTHDC1 expression activates DNA damage response, including faster cell cycle recovery, apoptosis evasion, and enhanced DNA repair capability.

Summary: Topoisomerase IIIa is a type 1A topoisomerase that forms a complex with RMI1 and RMI2 called TRR in human cells. In this study, the authors used optical tweezers and fluorescence microscopy to investigate the catalytic steps of TRR. They found that TRR forms an open gate in single-stranded DNA and can bind a second single-stranded or double-stranded DNA molecule, leading to catenation. Interestingly, double-stranded DNA binding increases the gate size and the helicase BLM affects the flexibility of the gate. These findings suggest that TRR-mediated transfer of double-stranded DNA may be more important than previously believed.

Summary: Cellular senescence is a powerful mechanism for inhibiting the proliferation of cancer cells, and RNA-binding proteins (RBPs) play important roles in regulating senescence. This study discovered that the RBP RBM4 contributes to cellular senescence and its depletion induces senescence in various cell types, including cancer cells. Furthermore, the depletion of RBM4 inhibits cancer cell progression and is mediated by the regulation of SERPINE1 through miR-1244.

Summary: This study identified a wheat RNA-binding protein 1 (TaRBP1) that acts as a negative regulator in response to infection by Puccinia striiformis f. sp. tritici (Pst). Knockdown of TaRBP1 resulted in enhanced resistance to Pst, suggesting its role in plant defence. TaRBP1 interacted with TaGLTP and prevented its degradation in plants. The findings reveal a novel susceptible mechanism by which plants fine-tune their defence responses during Pst infection.
