Summary: We report a new positively charged azidoamino acid for strain-promoted azide-alkyne cycloaddition (SPAAC) applications that overcomes the solubility limitations of commonly used azidolysine, especially in systems with numerous ligation sites. The synthesized residue is compatible with Fmoc-based solid-phase peptide synthesis and exhibits efficient second-order rate constants in SPAAC ligations employing DBCO (0.34 M-1 s-1) and BCN (0.28 M-1 s-1).

Summary: This study reports a novel antimicrobial peptide (A3P7) with potent antibacterial activity and moderate hemolytic activity. C-terminal truncation resulted in eight derivatives with reduced hemolytic activity while maintaining antibacterial activity. Among them, AP19 exhibited the best combination of broad-spectrum antibacterial activity and minimal hemolytic and cytotoxic activities. A D-form derivative (D-AP19) showed maintained antibacterial activity and potent activity against multi-drug resistant Acinetobacter baumannii. It displayed rapid bacterial killing via membrane disruption and had a low tendency to induce bacterial resistance.

Summary: A Pd-catalyzed C(sp(2))-H acetoxylation method for Tyr-containing peptides is described, utilizing a removable 2-pyridyloxy group as a directing group. This method offers scalable and easily tunable regioselectivity for mono- and diacetoxylation reactions. Interestingly, the formation of L-DOPA peptidomimetics is hindered upon removal of the directing group.
