Divergent Synthesis of Substituted Amino-1,2,4-triazole Derivatives

Summary: An efficient assembly of disubstituted 1,2,4-triazoles was achieved by cyclization of readily accessible N'-nitro-2-hydrocarbylidene-hydrazinecarboximidamides with moderate to excellent yields under mild reaction conditions. This divergent synthetic strategy allows for the formation of amino-1,2,4-triazoles under acidic conditions and nitroimino-1,2,4-triazoles under neutral conditions with water as the solvent.

Mechanochemical Synthesis of 1,2,4-Triazoles via a [3+2] Cycloaddition of Azinium-N-Imines and Nitriles

Summary: A mechanochemical Cu-catalyzed [3 + 2] cycloaddition of azinium-N-imines with nitriles was reported under solventless grinding conditions. Various 1,2,4-triazolos derivatives were obtained in high yields. The developed protocol offers advantages in functional-group compatibility, scalability, and shorter reaction time.

Summary: This study reports the design, synthesis, and characterization of a series of 3,5-diphenyl-1,2,4-triazole substituted [1,2,4]triazolo[3,4-b][1,3,4]thiadiazole derivatives as selective hMAO-B inhibitors. Compound 42e and 42h exhibited the highest inhibitory activity against hMAO-B.

Summary: Novel annulated azaheterocycles have been synthesized, showing antimicrobial activity against Staphylococcus aureus and potentially exerting antibiotic effects through DNA gyrase inhibition. 4,4′-Dinitrimino-5,5′-diamino-3,3′-azo-bis-1,2,4-triazole: A High-Performing Zwitterionic Energetic Material

Summary: The novel energetic material DNDAABT was synthesized via mixed acid nitration of electrochemically generated TAABT, with various energetic salts also prepared and characterized. Despite predictions of low sensitivities, DNDAABT and its salts showed primary explosive sensitivities with relatively high thermal stabilities for primary N-nitramines.

Summary: A modular synthesis of highly substituted 3-azapyrroles has been developed using a three-step sequence comprising copper-catalyzed alkyne-azide cycloaddition, Rhodium(II)-acetate-promoted 1-ST denitrogenation, and cyclodehydration. The method proved to be highly efficient and tolerated a wide range of substituents.

Unravelling the anticancer potency of 1,2,4-triazole-N-arylamide hybrids through inhibition of STAT3: synthesis and in silico mechanistic studies

Summary: The study focused on the design, synthesis, pharmacokinetic profiles, and in vitro anticancer activity of novel C3-linked 1,2,4-triazole-N-arylamide hybrids for inhibition of STAT3. Four derivatives showed promising cytotoxic effects against selected cancer cells, with compound 12 exhibiting the highest potency. These compounds were analyzed for their potential as drug candidates based on their ADMET properties.

This method does not require the use of transition metal catalysts and can be performed on a gram scale.

Tandem Cycloaddition of Azides to 3,3-Diaminoacrylonitriles (2-Cyanoacetamidines) and Cornforth-Type Rearrangement as an Approach to 5-Amino-1,2,3-triazole-4-N-substituted Imidamides

Summary: An efficient metal-free method for synthesizing 5-amino-1,2,3-triazole-4-N-sulfonyl- and arylimidamides has been developed based on a tandem process involving cycloaddition reaction and Cornforth-type rearrangement. The reaction optimization identified several factors that facilitate the reaction, including the use of a strong base and specific 3,3-diaminoacrylonitriles. The method has a broad scope and can be applied to obtain various 5-amino-1,2,3-triazole-4-carbimidamides with different substituents.

Base-Induced Highly Regioselective Synthesis of N2-Substituted 1,2,3-Triazoles under Mild Conditions in Air

Summary: This study developed a highly regioselective synthesis of N-2-substituted 1,2,3-triazoles using N-sulfonyl-1,2,3-triazoles and alkyl bromides/alkyl iodides at room temperature. The mechanistic pathway proposed explains the high N-2 regioselectivity, and the protocol showed good to excellent product yields (72-90%).

Synthesis and Antitumor Activity of 1-Substituted 1,2,3-Triazole-Mollugin Derivatives

Summary: A new series of mollugin-1,2,3-triazole derivatives were synthesized using a copper(I)-catalyzed Huisgen 1,3-dipolar cycloaddition reaction, and most of them demonstrated cytotoxicity on five human cancer cell lines, with compounds 14 and 17 showing significant cytotoxicity across all five cancer cell lines.

Facile Synthesis of Fully Substituted 1,2,4-Triazoles via [3+2] Cycloaddition of Nitrileimines with Amidine under Transition Metal-Free Conditions

Summary: An efficient method for the synthesis of densely 1,2,4-triazoles by [3+2] cycloaddition of nitrileimides with amidine hydrochlorides under transition metal-free conditions is reported. The method demonstrates mild reaction conditions, wide substrate scope, and good functional group tolerance. Various aryl, heterocyclic, and alkyl substituted 3-CF3-1,2,4-triazoles were synthesized with high yields up to 96% using NaHCO3 as a promoter. Additionally, the scalability and synthetic transformations of this method were successfully carried out, highlighting its utility.

Design and synthesis of new 2-oxoquinoxalinyl-1,2,4-triazoles as antitumor VEGFR-2 inhibitors

Summary: This study designed and synthesized a series of novel compounds as potential antitumor agents and VEGFR-2 inhibitors. Eight compounds showed high growth inhibition against MCF-7 cells and possessed sub-micromolar range of VEGFR-2 inhibitory activity. Docking studies and in silico physicochemical predictions suggested promising drug-likeness profiles for these compounds.
