Summary: Accumulating research has shown that excessive activation of microglia can lead to neuroinflammation and neurodegenerative diseases. This study investigated the effects of a compound called notopterol (NOT) on neuroinflammation and its underlying mechanisms. The findings demonstrated that NOT reduced the expression of pro-inflammatory mediators in cells exposed to LPS and activated the AKT/Nrf2/HO-1 signaling pathway. The anti-inflammatory effect of NOT was inhibited by specific inhibitors of AKT, Nrf2, and HO-1. Furthermore, NOT was able to protect BV-2 cells from LPS-induced damage and increase their survival rate. These results suggest that NOT inhibits the inflammatory response and exerts a neuroprotective effect by modulating the AKT/Nrf2/HO-1 signaling axis in microglia.

Activation of JAK2/STAT3 triggers chronic pain by interacting with microglia and astrocytes, releasing proinflammatory cytokines, inhibiting anti-inflammatory cytokines, and affecting synaptic plasticity. Retrospective analysis of pharmacological inhibitors for JAK2/STAT3 reveals their promising effect in treating different types of chronic pain.

Summary: This study demonstrated the therapeutic effects of dex in EAE by delaying clinical signs, motor defects, myelin and axon degeneration, immune cell infiltration, and neuroinflammatory activation in the early stages of the disease. Furthermore, the study highlighted the potential of mesencephalic astrocyte-derived neurotrophic factor (MANF) as a drug candidate for MS by showing its increased expression in dex-treated EAE mice and the improvement in clinical signs and motor behavior following hMANF administration. The time-dependent analysis provided insights into the disease progression and the link between MANF and EAE.

Summary: Amyotrophic lateral sclerosis (ALS) is a progressive neurodegenerative disease that affects motor neurons and has no cure or effective therapy. This study investigates the therapeutic effect of cerebral dopamine neurotrophic factor in rodent models of ALS and shows that it significantly halts disease progression and improves motor behavior. Cerebral dopamine neurotrophic factor rescues motor neurons from endoplasmic reticulum stress-related cell death and enhances motor neuron survival.
