Summary: CAPS1 is a key molecule involved in vesicular exocytosis, but its role in synaptic plasticity and learning behavior remains unclear. Studies showed impaired synaptic plasticity and learning behavior in Caps1 cKO mice, with reduced docked vesicles in the hippocampal CA3 region and impaired long-term potentiation in CA1 and CA3 regions. These findings suggest that CAPS1 plays a crucial role in hippocampal synaptic release and plasticity, which is essential for hippocampus-associated learning.

Summary: The study found that reduced expression of habenular CAPS2 in depression animal models and major depressive disorder patients, as well as knockdown of CAPS2, led to despair-like behavior, decreased release of DCVs in the IPN, and reduced neuronal activity in the IPN and monoaminergic centers. These results suggest that MHb CAPS2 plays a crucial role in depressive behavior through regulation of neuropeptide secretion and activity of monoaminergic centers in the MHb-IPN pathway.

Summary: Activation of voltage-gated calcium channels at presynaptic terminals triggers the fusion of synaptic vesicles, leading to neurotransmitter release. Caenorhabditis elegans neuromuscular junctions demonstrate the involvement of two classes of calcium channels, CaV2 and CaV1, in releasing distinct pools of synaptic vesicles. CaV2 channels are concentrated in dense clusters and mediate fusion of vesicles docked within a short distance, while CaV1 channels are dispersed throughout the synaptic varicosity and their vesicle fusion is dependent on a specific receptor.

Summary: This study investigated the endocytosis and replenishment of the immediately releasable pool (IRP) in mice primary chromaffin cells. The researchers found that not only a fast dynamin-dependent endocytosis occurred after the release of about 50% IRP, but also a fast dynamin-independent endocytosis was activated after depolarizations lasting 25-50ms, which released over 80% of IRP. This dynamin-independent endocytosis was found to be Ca2+- and protein kinase C (PKC)-dependent. Additionally, PKC inhibition also affected the slow component of replenishment observed after total IRP exocytosis.
