MicroRNA126 contributes to granulocyte colony-stimulating factor-induced hematopoietic progenitor cell mobilization by reducing the expression of vascular cell adhesion molecule 1

Background Mobilization of hematopoietic stem/progenitor cells from the bone marrow to the peripheral blood by granulocyte colony-stimulating factor is the primary means to acquire stem cell grafts for hematopoietic cell transplantation. Since hematopoietic stem/progenitor cells represent a minority of all blood cells mobilized by granulocyte colony-stimulating factor, the underlying mechanisms need to be understood in order to develop selective drugs. Design and Methods We analyzed phenotypic, biochemical and genetic changes in bone marrow cell populations from granulocyte colony-stimulating factor-mobilized and control mice, and linked such changes to effective mobilization of hematopoietic stem/progenitor cells. Results We show that granulocyte colony-stimulating factor indirectly reduces expression of surface vascular cell adhesion molecule 1 on bone marrow hematopoietic stem/progenitor cells, stromal cells and endothelial cells by promoting the accumulation of microRNA-126 (miR126)-containing microvescicles in the bone marrow extracellular compartment. We found that hematopoietic stem/progenitor cells, stromal cells and endothelial cells readily incorporate these miR126-loaded microvescicles, and that miR126 represses vascular cell adhesion molecule 1 expression on bone marrow hematopoietic stem/progenitor cells, stromal cells and endothelial cells. In line with this, miR126-null mice displayed a reduced mobilization response to granulocyte colony-stimulating factor. Conclusions Our results implicate miR126 in the regulation of hematopoietic stem/progenitor cell trafficking between the bone marrow and peripheral sites, clarify the role of vascular cell adhesion molecule 1 in granulocyte colony-stimulating factor-mediated mobilization, and have important implications for improved approaches to selective mobilization of hematopoietic stem/progenitor cells.

Summary: The differentiation of innate lymphoid cells (ILCs) from hematopoietic stem cells requires the progression through several multipotent progenitor stages. This study shows that there are distinct accessible chromatin regions in all lymphoid progenitors (ALPs), early ILC precursors (EILPs), and ILC precursors (ILCPs). Single-cell MNase-seq analyses reveal that EILPs contain subpopulations epigenetically primed toward dendritic cell or ILC lineages. Key transcription factors TCF-1 and GATA3 are found to be indispensable for the epigenetic priming of lineage-defining sites for ILCs at the EILP stage.

Summary: CD4 T helper (Th) cell subsets and their innate counterparts innate lymphoid cell (ILC) subsets display similar effector cytokine-producing capabilities during immune responses. The differentiation, development, and functions of these lymphoid cell subsets are regulated by lineage-determining transcription factors (LDTFs). This review discusses the functions of cis-regulatory elements in inducing LDTF expression during Th cell differentiation and ILC development, as well as the upstream signals involved in LDTF induction in vitro and in vivo. The possible mechanisms and physiological importance of regulating LDTF dynamic expression during ILC development and activation are also explored.

Summary: Interferon-y (IFN-y) is a key cytokine in response to viral or intracellular bacterial infection in mammals. A silencer (CNS-28) for the Ifng gene has been identified, which maintains Ifng silence by diminishing enhancer-promoter interactions within Ifng locus in a GATA3-dependent but T-bet-independent manner. CNS-28 plays a role in restraining Ifng transcription in NK cells, CD4+ cells, and CD8+ T cells, and its deficiency results in repressed type 2 immune responses and a shift in Th1 and Th2 paradigm.

HTR2A agonists play a therapeutic role by restricting ILC2 activation in papain-induced lung inflammation

Summary: This study found a negative correlation between the neurotransmitter serotonin and the cytokines IL-5 and IL-13 produced by ILC2s, which are related to depression. Serotonin can alleviate lung inflammation by suppressing ILC2 activation. The serotonin receptor HTR2A is highly expressed on ILC2s and can be targeted with an agonist called DOI to inhibit ILC2 activation and reduce type 2 immune response. Targeting HTR2A on ST2(+) ILC2s could be a potential treatment strategy for type 2 lung inflammation.

Multiple tolerance checkpoints restrain affinity maturation of B cells expressing the germline precursor of a lupus patient-derived anti-dsDNA antibody in knock-in mice

Summary: Anti-dsDNA antibodies are a marker of systemic lupus erythematosus and their high-affinity reactivity is believed to be the result of somatic hypermutation. This study investigated whether low-affinity precursors of these antibodies are subjected to immune tolerance. The researchers generated a mouse model carrying germline-reverted sequences of high-affinity anti-dsDNA antibodies and found that multiple tolerance checkpoints prevented these low-affinity precursors from undergoing clonal expansion and affinity maturation in germinal centers.
