Molecular cloning, characterization and expression analysis of interferon-beta promoter stimulator 1 (IPS-1) gene from grass carp Ctenopharyngodon idella

IPS-1 (interferon-beta promoter stimulator 1), also known as MAVS/VISA/Cardif, plays a central role in antiviral immunity. In this manuscript, we cloned and characterized IPS-1 from grass carp Ctenopharyngodon idella (designated as CiIPS-1). The CiIPS-1 cDNA is 2412 bp long and consists of a 5' untranslated region (UTR) of 124 bp, a 3' UTR of 497 bp with three cytokine RNA instability motifs (ATTTA) and a polyadenylation signal (AATAAA), and an open reading frame (ORF) of 1791 bp encoding a polypeptide of 596 amino acids with a calculated molecular mass of 64.1 kDa and a theoretical isoelectric point of 4.79. Structural analysis showed that the CiIPS-1 protein contained an N-terminal CARD (caspase activation and recruitment domain), a central proline-rich domain, a putative TRAF2-binding motif and a C-terminal transmembrane domain. Similarity analysis of the deduced amino acid sequence of the CiIPS-1 by MatGAT software revealed that the CiIPS-1 shared 27.8-76.4% identity and 47.4-85.2% similarity with other known piscine IPS-1 sequences. The CiIPS-1 mRNA was constitutively expressed in the examined tissues, higher in spleen, and was induced by grass carp reovirus (GCRV) injection by semi-quantitative RT-PCR assay. Quantitative real-time RT-PCR analysis revealed that the CiIPS-1 mRNA expression was rapidly and significantly up-regulated in vivo and in vitro after GCRV infection, and the CiIPS-1 transcripts were also significantly enhanced in vitro post the synthetic double stranded RNA polyinosinic polycytidylic potassium salt (poly (I:C)) stimulation. These results indicated that CiIPS-1 was an inducible acute-phase protein and involved in the immune reaction to GCRV in grass carp. (C) 2010 Elsevier Ltd. All rights reserved.

Summary: In this study, a recombinant Lactobacillus casei expressing multiple fragments of the key outer capsid protein of grass carp reovirus (GCRV) was constructed, and an oral microcapsule live vaccine (SCL-56310-500) was prepared. The results showed that this vaccine effectively enhanced the immune response and suppressed GCRV replication in fish, leading to a significantly higher survival rate.

Summary: This review discusses the unique biological effects of IL-2 family cytokines in regulating differentiation, survival, and activation of multiple lymphocyte lineages, as well as their expression profiles, receptors, signaling transductions, and functions in teleosts. The IL-2 family cytokines in teleosts show relatively conserved functions in lymphocyte proliferation, differentiation, development, and inflammation, similar to their counterparts in mammals.

Chitooligosaccharide boosts the immunity of immunosuppressed blunt snout bream against bacterial infections

Summary: This study reveals that chitooligosaccharide (COS) has the ability to reverse cortisol-induced immunosuppression in fish and enhance the immune activity of macrophages. Oral administration of COS directly absorbed through the intestine significantly improves the innate immunity of blunt snout bream, leading to increased survival and reduced tissue damage. Thus, COS offers potential strategies for immunosuppression prevention and control in fish.

VP4/VP56/VP35 Virus-like Particles Effectively Protect Grass Carp (Ctenopharyngodon idella) against GCRV-II Infection

Summary: This study developed a prophylactic grass carp reovirus (GCRV) vaccine using virus-like particle (VLP) assembly technology, and the highly immunogenic candidates in the vaccine were VP4, VP56, and VP35. Expression of VP4, VP56, and VP35 in both Escherichia coli and Pichia pastoris systems resulted in successful assembly of uniform, stable, and non-toxic VP4/VP56/VP35 VLPs. Vaccination with P. pastoris VLPs significantly improved survival rate, IgM levels, tissue viral load, and antioxidant immune-related indicators in grass carp.

Temperature-regulated type II grass carp reovirus establishes latent infection in Ctenopharyngodon idella brain

Summary: This study investigated the latent infection of GCRV-II in asymptomatic grass carp with GCRV infection or exposure history. It was found that GCRV-II was detectable only in the brain of grass carp during latent infection, unlike the multi-tissue distribution observed in natural infection. Additionally, GCRV-II distribution in grass carp was notably affected by ambient temperature. This study provides insights into the mechanisms of GCRV-II latent infection and reactivation, contributing to the prevention and control of GCRV pandemics.

Vitamin D3 can effectively and rapidly clear largemouth bass ranavirus by immunoregulation

Summary: This study aimed to investigate the potential effectiveness of VD3 in treating Largemouth bass ranavirus (LMBV). The results showed that feeding Largemouth bass with 15000 IU/Kg and 20000 IU/Kg of VD3 significantly suppressed and cleared LMBV pathogens. In addition, VD3 treatment increased the expression levels of key immune-related genes and enhanced antioxidant and immune activities in the serum. Tissue pathology sections also revealed a dose-dependent relationship between VD3 supplementation and tissue damage.

Re-identification and characterization of grass carp Ctenopharyngodon idella TLR20
