LGP2 (laboratory of genetics and physiology 2), a homologue of RIG-I (Retinoic acid inducible gene-I) and MDA5 (Melanoma differentiation associated gene 5) without the CARD (caspase activation and recruitment domain) required for signaling, plays a pivotal role in modulating signaling by RIG-I and MDA5 for interferon (IFN) synthesis. In this study, a novel LGP2 gene from grass carp Ctenopharyngodon idella (designated as CiLGP2) was isolated and characterized. The full-length cDNA of CiLGP2 was of 2920 bp with five instability motifs (ATTTA). The open reading frame was of 2043 bp and encoded a polypeptide of 680 amino acids, including five main overlapping structural domains: two DEXDc (DEAD/DEAH box helicase domain), one Resin (conserved restriction domain of bacterial type III restriction enzyme), one HEL1Cc (helicase superfamily c-terminal domain) and one RD (regulatory domain). There was one more alpha-helix in the RD, compared with that in human. The CiLGP2 mRNA was ubiquitous expression in the tested tissues, was high level in spleen, skin, heart and intestine tissues, and was up-regulated by grass carp reovirus (GCRV) injection by semi-quantitative RT-PCR (sqRT-PCR) assay. The CiLGP2 expression in spleen was significantly up-regulated at 12 h (14.5 folds, P < 0.05), reached the crest at 24 h (19.0 folds, P < 0.05), and then dropped a little at 48 h (10.4 folds) post-injection of GCRV and kept this level in the following test period (P < 0.05). In liver, the temporal expression of CiLGP2 mRNA was significantly increased at 24 h (3.8 times, P < 0.05), reached peak at 48 h (10.7 times, P < 0.05), and then decreased a little bit at 72 h (5.8 times, P < 0.05) and kept this high level by the end of the test (P < 0.05). These results collectively suggested that CiLGP2 was a novel member of RLR gene family, engaging in the early stage of antiviral innate immune defense in grass carp, and laid the foundation for the further mechanism research of LGP2 in fishes. Jiao Chang, Wentao Zhu, Xingchen Huo, Meihua Qiao, Chunrong Yang, Yongan Zhang, Jianguo Su

Summary: In this study, a recombinant Lactobacillus casei expressing multiple fragments of the key outer capsid protein of grass carp reovirus (GCRV) was constructed, and an oral microcapsule live vaccine (SCL-56310-500) was prepared. The results showed that this vaccine effectively enhanced the immune response and suppressed GCRV replication in fish, leading to a significantly higher survival rate.

Interleukin-2 family cytokines: An overview of genes, expression, signaling and functional roles in teleost

Yanqi Zhang, Jianguo Su

Summary: This review discusses the unique biological effects of IL-2 family cytokines in regulating differentiation, survival, and activation of multiple lymphocyte lineages, as well as their expression profiles, receptors, signaling transductions, and functions in teleosts. The IL-2 family cytokines in teleosts show relatively conserved functions in lymphocyte proliferation, differentiation, development, and inflammation, similar to their counterparts in mammals.

Summary: This study evaluates the feasibility and biological activity of carboxymethyl chitosan (CMCS) loaded with Ctenopharyngodon idella interferon-gamma 2 (CiIFN-gamma 2), and examines the resistance to Aeromonas hydrophila infection in grass carp. The results show that CMCS-CiIFN-gamma 2 nanoparticles can enhance the expression of pro-inflammatory cytokines and the effective uptake of the protein, leading to improved immune protection against bacterial diseases in aquaculture.

Chitooligosaccharide boosts the immunity of immunosuppressed blunt snout bream against bacterial infections

Aotian Ouyang, Mengwei Zhang, Gailing Yuan, Xiaoling Liu, Jianguo Su

Summary: This study reveals that chitooligosaccharide (COS) has the ability to reverse cortisol-induced immunosuppression in fish and enhance the immune activity of macrophages. Oral administration of COS directly absorbed through the intestine significantly improves the innate immunity of blunt snout bream, leading to increased survival and reduced tissue damage. Thus, COS offers potential strategies for immunosuppression prevention and control in fish.

Type II Grass Carp Reovirus Rapidly Invades Grass Carp (Ctenopharyngodon idella) via Nostril-Olfactory System-Brain Axis, Gill, and Skin on Head

Wentao Zhu, Meihua Qiao, Meidi Hu, Xingchen Huo, Yongan Zhang, Jianguo Su

Summary: This study found that Type II grass carp reovirus (GCRV-II) rapidly invades grass carp through the nostril, gill, and skin on the head, rather than merely by adhesion. Further investigations revealed that GCRV-II invades grass carp via the nostril-olfactory system-brain axis, and viral replication occurs.

VP4/VP56/VP35 Virus-like Particles Effectively Protect Grass Carp (Ctenopharyngodon idella) against GCRV-II Infection

Qingqing Tian, Xingchen Huo, Qian Liu, Chunrong Yang, Yongan Zhang, Jianguo Su

Summary: This study developed a prophylactic grass carp reovirus (GCRV) vaccine using virus-like particle (VLP) assembly technology, and the highly immunogenic candidates in the vaccine were VP4, VP56, and VP35. Expression of VP4, VP56, and VP35 in both Escherichia coli and Pichia pastoris systems resulted in successful assembly of uniform, stable, and non-toxic VP4/VP56/VP35 VLPs. Vaccination with P. pastoris VLPs significantly improved survival rate, IgM levels, tissue viral load, and antioxidant immune-related indicators in grass carp.

Temperature-regulated type II grass carp reovirus establishes latent infection in Ctenopharyngodon idella brain

Rui Jiang, Jie Zhang, Zhiwei Liao, Wentao Zhu, Hang Su, Yongan Zhang, Jianguo Su

Summary: This study investigated the latent infection of GCRV-II in asymptomatic grass carp with GCRV infection or exposure history. It was found that GCRV-II was detectable only in the brain of grass carp during latent infection, unlike the multi-tissue distribution observed in natural infection. Additionally, GCRV-II distribution in grass carp was notably affected by ambient temperature. This study provides insights into the mechanisms of GCRV-II latent infection and reactivation, contributing to the prevention and control of GCRV pandemics.

Vitamin D3 can effectively and rapidly clear largemouth bass ranavirus by immunoregulation

Pengxu Wang, Xingchen Huo, Fengxia Zhao, Qian Liu, Qingqing Tian, Chunrong Yang, Jianguo Su

Summary: This study aimed to investigate the potential effectiveness of VD3 in treating Largemouth bass ranavirus (LMBV). The results showed that feeding Largemouth bass with 15000 IU/Kg and 20000 IU/Kg of VD3 significantly suppressed and cleared LMBV pathogens. In addition, VD3 treatment increased the expression levels of key immune-related genes and enhanced antioxidant and immune activities in the serum. Tissue pathology sections also revealed a dose-dependent relationship between VD3 supplementation and tissue damage.
