The Golgi apparatus: Lessons from Drosophila

Historically, Drosophila has been a model organism for studying molecular and developmental biology leading to many important discoveries in this field. More recently, the fruit fly has started to be used to address cell biology issues including studies of the secretory pathway, and more specifically on the functional integrity of the Golgi apparatus. A number of advances have been made that are reviewed below. Furthermore, with the development of RNAi technology, Drosophila tissue culture cells have been used to perform genome-wide screens addressing similar issues. Last, the Golgi function has been involved in specific developmental processes, thus shedding new light on the functions of a number of Golgi proteins. Summary: In the absence of autophagy defects, activation of p62 and Nrf2 exacerbates liver disease phenotype and can lead to early death. Expression of a p62 mutant exacerbates hepatocarcinogenesis, while forced activation of Nrf2 itself does not increase liver injury or tumor burden.

ADAR1 averts fatal type I interferon induction by ZBP1

Summary: Mutations of the ADAR1 gene cause chronic activation of type I interferon responses, leading to severe diseases. The interaction between ADAR1 and another protein called ZBP1 prevents IFN activation. Deficiency or mutation of ZBP1's Z alpha domains reduces expression of IFN-stimulated genes and prevents early postnatal lethality in mice with mutated ADAR1 Z alpha domain.

Summary: Organoids hold great promise for our understanding of organ development and disease study, but there is heterogeneity among organoids. This study reveals that batch-to-batch variation is low, but passage number has a profound impact on gene expression profiles. Additionally, organoid-to-organoid variability exists within a culture, which may be influenced by the microenvironment.

Summary: This study found that activated hepatic stellate cells induce autophagy, leading to the release of miR-29a. Inhibition of autophagy can suppress fibrogenic gene expression by reducing the release of miR-29a.
