Summary: This study identified two gene signatures capturing intrinsic and extrinsic features in melanoma using TCGA RNA-seq data. By decomposing gene expression into representative nodes, the study found nodes significantly associated with patient prognosis, leading to the definition of tumor-intrinsic and tumor-extrinsic signatures. These signatures were able to predict patient prognosis and were positively associated with patient response to immunotherapies.

Various types of vaccines, such as DNA, peptide, protein, dendritic cell-based, and endothelial cell vaccines, have been studied to target angiogenesis in melanoma for potential progress in developing effective treatments.

Summary: This study explores an intensive tumor immunotherapeutic strategy by combining mannosylated nanovaccines with gene regulated PD-L1 blockade. The mannose modified nanovaccines can effectively co-encapsulate antigen and adjuvant, promoting endocytosis and cross presentation in dendritic cells. However, the limited inhibition of tumor growth by nanovaccines in the immunosuppressed tumor microenvironment can be overcome by combining with gene regulated PD-L1 blockade, leading to significant tumor remission.

Summary: In this study, immune-related lncRNAs were correlated with patient survival in melanoma, leading to the development of a prognostic model. Through multivariate Cox regression analyses, 8 immune-related lncRNAs were identified as having prognostic value, and a predictive model based on these variables effectively predicted patient prognosis. Additionally, significant differences in the distribution of immune-related genes were observed between high and low-risk groups, with higher immune score and immune function enrichment score in the low-risk group.
