Summary: The complex and costly manufacturing process of CAR-T cell therapy has hindered its widespread adoption. Induced pluripotent stem cells (iPSCs) have potential as an alternative cellular immunotherapy due to their unlimited self-renewal potential and ability to differentiate into functional immune cells. Careful selection of the original cell for iPSC seed preparation is crucial due to the presence of epigenetic imprint. Our research shows that lymphocyte-origin cells are superior to embryonic stem cells in differentiating into functional immune cells. Blocking Fas-FasL induced apoptosis in T cells significantly improves iPSC generation without affecting their differentiation potential.

This study provides evidence for the feasibility of regenerative medicine strategies using clinically relevant materials in the treatment of severe chronic SCI.

Summary: The study demonstrates that transplantation of aligned cardiac tissue derived from iPS cells improves cardiac function with angiogenesis and antifibrotic effects in a porcine cardiomyopathy model. In both in vitro and in vivo studies, the aligned cardiac tissue containing high purity cardiomyocytes showed homogeneous contraction and excellent mechanical properties, leading to significant improvement in left ventricular ejection fraction compared to the control group. Limited engraftment of the transplanted tissue was observed, but the therapeutic effects were still prominent.

Additionally, the interactions between CAR-T cells and the host and tumor microenvironment significantly impact their activity. Therefore, innovative methodologies are needed to engineer more potent CAR-T cells with greater anti-tumor activity and less toxicity.
