Summary: Parkinson's disease (PD) is a neurodegenerative disorder associated with factors like oxidative stress, inflammation, and mitochondrial dysfunction. Ca2+ signaling plays a key role in PD and other neuronal diseases. This study found that apamin (APM), a selective antagonist of the SK channel, can suppress neuroinflammatory response. The researchers discovered that APM reduces Ca2+ overload from MPP+-induced neurotoxicity by regulating CaMKII, ERK, and NF kappa B / STAT3 signaling pathways. This suggests that APM may prevent loss of dopaminergic neurons through inhibition of Ca2+-overload-mediated signaling pathway.

Summary: This study found that patients with atrial fibrillation (AF) have higher SK-channel current (I-SK) in their heart cells, and the antiarrhythmic drug apamin can significantly prolong the action potential of these cells. In addition, the study also found that the abundance of SK2 channel on the cell membrane is higher in AF patients. It was discovered that inhibiting the phosphatase-2A-dependent calmodulin-Thr80 dephosphorylation and promoting the trafficking and targeting of SK-channel subunits can enhance the function of SK channels.

Summary: This study investigates the strategy of equipping low-molecular weight targeted radiotherapeutics with irreversible cysteine cathepsin inhibitors to increase tumor retention and therapeutic dose delivery. By incorporating ETs with different charge states into NTSR1-targeted antagonists, the impact on inhibition kinetics and adduct formation was examined, with minimal effects on NTSR1 targeting but increased renal uptake observed. Further exploration of the endolysosomal trapping approach in receptor-targeted antagonistic constructs is warranted based on the results.

Summary: This study aimed to design and develop different dosage forms of nifedipine/indomethacin fixed-dose combination (FDC) to provide delayed-extended and immediate release profiles. The developed FDC dosage forms showed significant improvement in drug release profiles and remained stable during the stability study. These designed dosage forms could enhance patient compliance and improve the quality of life.

The syntheses of Ga-68-DOTA-NT and Lu-177-DOTA-NT showed high purity and stability, indicating the potential of neurotensin in delivering imaging/therapeutic Ga-68/Lu-177 radioisotopes pair. Further evaluation is needed to assess the possible radiotoxicity effects on the liver, kidneys, or bone marrow.

Summary: Ellagic acid has been studied for its potential in preventing and treating cancer. In this study, APA-functionalized emulsomes were used to enhance the cytotoxic activity of ellagic acid against breast cancer cells, resulting in significant inhibition of cell growth and induction of apoptosis.

Summary: The study demonstrated the presence of all three beta-adrenoceptor subtypes in the endothelium of rat cremaster muscle artery, with beta(3)-adrenoceptor mediated vasodilation only evident after blockade of beta(1/2)-adrenoceptors. Constitutive beta(1/2)-adrenoceptor activity inhibits beta(3)-adrenoceptor function in the endothelium of skeletal muscle resistance arteries.
