Synthesis, structures, and antimalarial activities of some silver(I), gold(I) and gold(III) complexes involving N-heterocyclic carbene ligands

A series of mono-and dinuclear silver(I) and mononuclear gold(I) complexes containing bis(N-hetero-cyclic carbene) (NHC) or N-functionalized NHC ligands were synthesized and fully characterized by spectroscopic methods and, in some cases, by single crystal X-ray diffraction. The in vitro antiplasmodial and antifungal activities of a previously described family of N-functionalized bis(imidazolium) proligands and their corresponding silver(I), gold(I) and gold(III) complexes but also the new here described compounds were investigated in a chloroquine-resistant strain of Plasmodium falciparum, and against two Candida strains, respectively. For the first family, interesting antiplasmodial and antifungal activities were found for the dinuclear silver(I) species but they also showed strong hemolytic properties. Pharmaco-modulations leading to the second series of complexes allowed notably increase in the antiplasmodial activity, in particular of the mononuclear gold(I) complexes with IC50 values up to. 330 nM, without any hemolysis. Superoxide: A major role in the mechanism of action of essential antimalarial drugs

Summary: The research found that artemisinins, atovaquone, and chloroquine can generate significant levels of superoxide radicals in Plasmodium-infected erythrocytes, while the production of superoxide was lowered in chloroquine-resistant strain of Plasmodium treated with chloroquine. Reactive Oxygen Species as the Brainbox in Malaria Treatment

Summary: Various measures are taken to combat the spread of malaria globally, with most common antimalarials utilizing ROS-mediated approaches to kill parasites, but varying levels of ROS generation can affect their pharmacological activities. The development of resistance by Plasmodium falciparum is a growing concern, possibly due to the parasites' ability to manage oxidative stress. Combining conventional drugs with ROS-generating ability with newer drugs targeting vital metabolic pathways, like antioxidant machinery, could be a way forward for effective malaria control.

Synthesis and Antimalarial Activities of New Hybrid Atokel Molecules

Summary: In response to the urgent need for new efficient drugs to combat the spreading resistance of malaria parasite Plasmodium falciparum to artemisinin-based combination therapies, a series of novel hybrid drugs named Atokels were synthesized and characterized. These drugs, based on an 8-amino- or 8-hydroxyquinoline entity covalently bound to a 1,4-naphthoquinone through a polyamine linker, have been designed to target the parasite mitochondrion and induce damaging oxidative stress. Promising antimalarial activity was shown by the most effective Atokel drug on an artemisinin-resistant P. falciparum strain, with no cytotoxicity at 50 μm.

A pro-death autophagy-based nanoplatform for enhancing antitumour efficacy with improved immune responses

Summary: This study proposes a novel nanoplatform based on the autophagy cascade to overcome the obstacles in chemo-immunotherapy. The platform combines chemotherapy and starvation therapy to initiate pro-death autophagy and enhance antigen presentation, while also remodeling the immunosuppressive tumor microenvironment. Furthermore, the study discovers a new therapeutic direction for the respiration inhibitor 3-bromopyruvic acid (3BP) in cancer treatment. Overall, this study offers an opportunity to improve antitumor efficacy and boost immune responses.

A novel scaffold long-acting selective estrogen receptor antagonist and degrader with superior preclinical profile against ER plus breast cancer

Summary: Breast cancer is a common malignant tumor in women, and drug resistance remains a clinical challenge. In this study, a novel compound, G-5b, was developed with potent antagonistic and degradation activities comparable to the current drug fulvestrant. G-5b also showed improved stability and solubility. Mechanistically, G-5b engages the proteasome pathway to degrade ER, inhibiting the ER signaling pathway and inducing apoptosis and cell cycle arrest. In animal models, G-5b exhibited superior pharmacokinetics and pharmacodynamics properties. Overall, G-5b is a promising long-acting SERD worthy of further investigation and optimization.

Summary: A series of hybrid inhibitors combining pharmacophores of known kinase inhibitors and benzohydroxamate HDAC inhibitors were synthesized and evaluated for their anticancer activity and pharmacokinetic properties. Compounds 4d-f exhibited promising cytotoxicity against hematological cells and moderate activity against solid tumor models. Compound 4d showed potent inhibition of multiple kinase targets and had stable interactions with HDAC and members of the JAK family. These compounds showed selective cytotoxicity with minimal effects on non-tumorigenic cells and favorable pharmacokinetic profiles.

Unexpected rearrangement of ivermectin in the synthesis of new derivatives with trypanocidal and antiplasmodial activities

Design and synthesis of pterostilbene derivatives bearing triazole moiety that might treat DSS-induced colitis in mice through modulation of NF-KB/ MAPK signaling pathways

Summary: In this study, a series of novel anti-inflammatory compounds were designed and synthesized based on the natural product pterostilbene skeleton. Among them, compound 8 showed the highest activity and exhibited its effects through inhibition of pro-inflammatory cytokines by blocking the NF-KB/MAPK signaling pathway. Compound 8 also demonstrated a good relieving effect on acute colitis in mice and showed good safety in acute toxicity experiments.

Discovery of 4-(N-dithiobenzyl piperazine)-1,8-naphthalimide as a potent multi-target antitumor agent with good efficacy, limited toxicity, and low resistance

Some compounds exhibited significant antitumor activity and one compound showed excellent efficacy, limited toxicity, and low resistance. Further mechanism studies revealed that the compound exerted antitumor effects through multiple pathways.
