A new class of anticonvulsants possessing 6 Hz activity: 3,4-Dialkyloxy thiophene bishydrazones

Thirty nine new 3,4-di(substituted)oxy-N-2,N-5-bis(substituted)thiophene-2,5-dicarbohydrazides were synthesized starting from ethyl thiodiglycolate through multi-step reactions. In the synthetic sequence, 3,4-dihydroxythiophene-2,5-diester (1) was obtained by condensing the ethyl thiodiglycolate with diethyl oxalate. It was derivatized using different alkyl halides to give disubstituted thiophene esters (25), which were then converted to corresponding hydrazides (6-9) following usual methods. Finally, these hydrazides, on treatment with various substituted carbonyl compounds underwent smooth condensation to yield target hydrazones (10-13). The new compounds were characterized using FT-IR, H-1 NMR and C-13 NMR, mass spectral and elemental analyses. The anticonvulsant activity of the title compounds was established after intraperitoneal (ip) administration in three seizure models, which include maximal electroshock (MES), subcutaneous pentylenetetrazole (scPTZ) and 6 Hz screens and their neurotoxicity was also evaluated. Compound 11f has emerged as an active compound with no neurotoxicity in this series. Also, the structure-activity relationship of the tested compounds was discussed. (C) 2009 Elsevier Masson SAS. Push-Pull Phenoxazine-Based Sensitizers for p-Type DSSCs: Effect of Acceptor Units on Photovoltaic Performance

Summary: Finding efficient p-type sensitizers for NiO photocathodes is a challenge for developing low-cost tandem dye-sensitized solar cells. In this study, six new organic dyes were designed and synthesized for sensitizing mesoporous p-type semiconductor NiO in p-type DSSCs. Optical studies showed that these dyes exhibited suitable absorption and emission features, with thermodynamic parameters favoring charge transfer involving NiO. One of the dyes achieved the highest power conversion efficiency in the devices.

New Carbazole-Based Sensitizers for p-Type DSSCs: Impact of the Position of Acceptor Units on Device Performance

Summary: We report the design, synthesis, and characterization of two new carbazole-based organic dyes PC1-2 as potential sensitizers for p-type dye-sensitized solar cells. PC1 exhibited better performance compared to PC2, with a higher power conversion efficiency of 0.027%.

New green emitters based on push-pull type pyrene substituted cyanopyridones: Design strategies and utilization in organic light-emitting diodes

Summary: A series of novel D-A-D type conjugated dyes (Py-CyP1-7) with a strongly electron-withdrawing cyano-pyridone scaffold (A) linked to highly electron-donating pyrene unit (D) and varied auxiliary donors (D) were designed and synthesized for green organic light-emitting diodes (OLEDs). The efficiency of these hybrids as green emitters was validated through theoretical, optical, electrochemical, and thermal studies. Finally, the dyes were used as sole and dopant emitters in OLEDs, and the OLED with 3 wt % Py-CyP4 in the CBP host exhibited maximum efficiencies of 14.38 cd A ¡1, 12.04 lm W ¡1, and 5.91%.

A pro-death autophagy-based nanoplatform for enhancing antitumour efficacy with improved immune responses

Summary: This study proposes a novel nanoplatform based on the autophagy cascade to overcome the obstacles in chemo-immunotherapy. The platform combines chemotherapy and starvation therapy to initiate pro-death autophagy and enhance antigen presentation, while also remodeling the immunosuppressive tumor microenvironment. Furthermore, the study discovers a new therapeutic direction for the respiration inhibitor 3-bromopyruvic acid (3BP) in cancer treatment. Overall, this study offers an opportunity to improve antitumor efficacy and boost immune responses.

A novel scaffold long-acting selective estrogen receptor antagonist and degrader with superior preclinical profile against ER plus breast cancer

Summary: Breast cancer is a common malignant tumor in women, and drug resistance remains a clinical challenge. In this study, a novel compound, G-5b, was developed with potent antagonistic and degradation activities comparable to the current drug fulvestrant. G-5b also showed improved stability and solubility. Mechanistically, G-5b engages the proteasome pathway to degrade ER, inhibiting the ER signaling pathway and inducing apoptosis and cell cycle arrest. In animal models, G-5b exhibited superior pharmacokinetics and pharmacodynamics properties. Overall, G-5b is a promising long-acting SERD worthy of further investigation and optimization.

Summary: A series of hybrid inhibitors combining pharmacophores of known kinase inhibitors and benzohydroxamate HDAC inhibitors were synthesized and evaluated for their anticancer activity and pharmacokinetic properties. Compounds 4d-f exhibited promising cytotoxicity against hematological cells and moderate activity against solid tumor models. Compound 4d showed potent inhibition of multiple kinase targets and had stable interactions with HDAC and members of the JAK family. These compounds showed selective cytotoxicity with minimal effects on non-tumorigenic cells and favorable pharmacokinetic profiles.

Unexpected rearrangement of ivermectin in the synthesis of new derivatives with trypanocidal and antiplasmodial activities

Design and synthesis of pterostilbene derivatives bearing triazole moiety that might treat DSS-induced colitis in mice through modulation of NF-KB/ MAPK signaling pathways

Summary: In this study, a series of novel anti-inflammatory compounds were designed and synthesized based on the natural product pterostilbene skeleton. Among them, compound 8 showed the highest activity and exhibited its effects through inhibition of pro-inflammatory cytokines by blocking the NF-KB/MAPK signaling pathway. Compound 8 also demonstrated a good relieving effect on acute colitis in mice and showed good safety in acute toxicity experiments.
