NordicDB: a Nordic pool and portal for genome-wide control data

A cost-efficient way to increase power in a genetic association study is to pool controls from different sources. The genotyping effort can then be directed to large case series. The Nordic Control database, NordicDB, has been set up as a unique resource in the Nordic area and the data are available for authorized users through the web portal (http://www.nordicdb.org). The current version of NordicDB pools together high-density genome-wide SNP information from similar to 5000 controls originating from Finnish, Swedish and Danish studies and shows country-specific allele frequencies for SNP markers. The genetic homogeneity of the samples was investigated using multidimensional scaling (MDS) analysis and pairwise allele frequency differences between the studies. The plot of the first two MDS components showed excellent resemblance to the geographical placement of the samples, with a clear NW-SE gradient. We advise researchers to assess the impact of population structure when incorporating NordicDB controls in association studies. This harmonized Nordic database presents a unique genome-wide resource for future genetic association studies in the Nordic countries. Genome-wide association study meta-analysis of suicide death and suicidal behavior

Summary: This study aimed to identify genetic risk variants associated with suicide death and suicidal behavior. The results showed that a locus in the neuroligin 1 (NLGN1) gene was associated with suicide death, while ROBO2 and ZNF28 genes were associated with suicidal behavior. Additionally, variants near SOX5 and LOC101928519 were found to be associated with suicidal attempts. Suicide death and suicidal behavior showed positive correlations with depression, schizophrenia, pain, and suicidal attempt, and negative correlation with educational attainment.

Summary: A study in Finland found that reducing the price of alcoholic beverages increases alcohol-related health problems. However, genetic susceptibility to alcohol consumption did not become more significant in predicting these problems before and after the price reform.

Summary: A new analysis of large biobanks reveals genetic variants associated with changes in medication for cardiometabolic diseases and presents polygenic scores of medication-use behavior. The study used data from the Finnish nationwide drug purchase registry and performed genome-wide association analyses to identify loci associated with medication use in hyperlipidemia, hypertension, and type 2 diabetes. The findings demonstrate the potential for medication-based strategies to identify cardiometabolic risk loci and provide genome-wide tools for preventing cardiovascular diseases.

Summary: Six rare SLCO1B1 single nucleotide variants (SNVs) were identified in Finnish individuals with a psychotic disorder, which may affect the expression and functionality of the OATP1B1 protein. Functional studies showed that one of these SNVs (c.629G>T) resulted in the loss of OATP1B1 function and decreased membrane protein expression.

Summary: Through a large-scale multi-ancestry genome-wide association meta-analysis, 1020 independent association signals implicating 559 genes related to COPD were identified. These genes were enriched in 29 pathways. Variants showed heterogeneity across ancestries, age and smoking groups, and the genetic risk score was strongly associated with COPD across ancestry groups. New potential causal variants, genes, proteins and pathways were highlighted, including those targeted by existing drugs. These findings contribute to a better understanding of lung function and COPD mechanisms and have implications for functional genomics experiments and future COPD therapies.

Use of electronic health record data mining for heart failure subtyping

Summary: This study aims to assess whether electronic health record data text mining can be used to improve register-based heart failure subtyping. The results show that quantitative ejection fraction data can be efficiently extracted from electronic health records and used with laboratory data to subtype heart failure with reasonable accuracy, especially for heart failure with reduced ejection fraction.

Genetic and environmental factors underlying parallel changes in body mass index and alcohol consumption: a 36-year longitudinal study of adult twins

Summary: This study examined the genetic and environmental factors underlying simultaneous changes in body weight and alcohol consumption. The results showed that both genetic and environmental factors play a role in the changes of these traits.
