The lymphatic system is indispensable for the collection and cycling of tissue-extravasated fluids, macromolecules and immune cells into the bloodstream. Different mechanisms, including sprouting, ballooning and budding of lymphatic endothelial cells from the cardinal vein, have been proposed for lymphatic vessel formation during mammalial embryogenesis. Hagerling et al (2013) now provide a cell-scale model of lymphoangiogenesis by applying selective plane illumination-based ultra-microscopy (Becker et al, 2008) to wholemount-immunostained mouse embryos. They describe VEGFR-3, VEGF-C and CCBE1 as key regulators of lymphatic endothelial cell budding and migration at the early emergence of lymphatics from venous endothelium.

TFEB inhibition induces melanoma shut-down by blocking the cell cycle and rewiring metabolism

Summary: Melanomas are characterized by dysregulation of MAPK pathway, glycolysis, and TCA cycle, and the oncogenic transcription factor EB (TFEB) controls melanoma tumor growth through regulating ERK1/2 activity and metabolism. TFEB negatively regulates DUSP-1, dephosphorylates ERK1/2, and inhibits melanoma cell proliferation. TFEB also influences glucose, glutamine, and cholesterol metabolism, affecting glycolysis, glutaminolysis, and oxidative phosphorylation. TFEB silencing impairs cholesterol synthesis, glucose and glutamine uptake, and reverses TCA cycle, leading to fatty acid production. In a melanoma model, TFEB silencing reduces tumor growth, increases DUSP-1 level, and inhibits ERK1/2 action, indicating the importance of TFEB in melanoma cell behavior and metabolic pathways.

Long Non-Coding RNA LINC02802 Regulates In Vitro Sprouting Angiogenesis by Sponging microRNA-486-5p

Summary: Recent evidence suggests that noncoding RNAs are important for regulating angiogenesis. This study identified a noncoding RNA called LINC02802 that plays a critical role in regulating capillary sprouting. LINC02802 acts as a competing endogenous RNA with miR-486-5p to regulate the expression of the Mastermind-like-3 gene.
