A new method based on partial filling-MEKC (PF-MEKC) directly coupled to ESI-MS was developed for the simultaneous separation and determination of 13 structurally similar cytokinins, including various geometric and positional isomers of cytokinins. On the basis of the resolution of the neighboring isomer peaks, different parameters (i.e., pH and concentration of buffer, surfactant concentrations, length of the injected micellar plug, organic modifier, and applied separation voltage) were optimized to achieve a satisfactory PF-MEKC separation. Under optimum conditions, the separation of 13 cytokinin standards was accomplished within 25 min. MS/MS with multiple reaction monitoring detection was carried out to obtain sufficient selectivity. PF-MEKC-MS/MS allowed for the direct identification and confirmation of the cytokinins present in banana (Musa spp.) pulp sample after extraction and purification. Finally, trans-zeatin riboside (ZR) and trans-zeatin (Z) were unambiguously identified in banana pulp. It is anticipated that the current PF-MEKC-MS method can be applied to analyze cytokinins in a wide range of biological samples.

Relaxin elicits renoprotective actions accompanied by increasing bile acid levels in streptozotocin-induced diabetic mice

Chen Huei Leo, Jamie Li Min Ou, Eng Shi Ong, Cheng Xue Qin, Rebecca H. Ritchie, Laura J. Parry, Hooi Hooi Ng

Summary: This study evaluates the protective effects of relaxin on diabetic kidney complications. The results show that relaxin treatment can reduce key markers of kidney fibrosis, oxidative stress, and inflammation in a diabetic mouse model, and increase the levels of bile acid metabolites. Therefore, relaxin may serve as an adjunctive treatment for diabetic kidney complications.

Recent advances in the combination of organic solvent-free extraction, chemical standardization, antioxidant assay, and cell culture metabolomics for functional food and its by-product

Chen Huei Leo, Eng Shi Ong

Summary: Functional foods and their by-products have various bioactive components that offer health benefits and can enhance public health and well-being. Environmentally friendly, sustainable, and cost-effective methods are needed to extract flavonoids, phenolic compounds, and other substances from functional food and agri-food wastes to achieve a circular economy. This research covers extraction methods that eliminate the use of organic solvents and are suitable for laboratory and commercial production. Analytical methods are used to determine marker or bioactive compounds and chemical fingerprints to control the composition variation of functional food products. Combining chemical standardization with antioxidant assays provides information on extract quality. Cellular models and other experimental endpoints, such as metabolomics, are used to validate the antioxidant properties of the extracts.

Metabolic Processes and Biological Macromolecules Defined the Positive Effects of Protein-Rich Biostimulants on Sugar Beet Plant Development

Okanlawon L. Jolayemi, Ali H. Malik, Ramesh R. Vetukuri, Ganapathi V. Saripella, Pruthvi B. Kalyandurg, Tobias Ekblad, Jean W. H. Yong, Marie E. Olsson, Eva Johansson

Summary: Protein-based biostimulants (PBBs) enhance plant development by upregulating genes related to protein and photosynthesis. Hydrolyzed wheat gluten (HWG) and potato protein film (PF) were effective PBBs for promoting sugar beet growth and improving protein-related traits. Sucrose accumulation in the roots was mainly influenced by the availability of nitrogen.

Pressurized Hot Water Extraction of Mangosteen Pericarp and Its Associated Molecular Signatures in Endothelial Cells

Sakeena Si Yu Tan, Meyammai Shanmugham, Yu Ling Chin, Jia An, Chee Kai Chua, Eng Shi Ong, Chen Huei Leo

Summary: This study extracts phytochemical compounds from mangosteen pericarp using pressurized hot water extraction (PHWE) and investigates its biological effects on endothelial cells using RNA sequencing. Results show that extraction at a temperature of 120 degrees Celsius yields the highest concentrations of key phytochemical compounds and improves antioxidant capacity. Treatment of endothelial cells with the extract activates the AMPK signaling pathway and affects biological processes related to steroid biosynthesis and metabolism. Overall, this study provides important insights into the potential beneficial effects of phytochemical compounds from mangosteen pericarp on cardiometabolic health.

Summary: This study explores the use of 3D printing technology to transform orange peel waste into edible, healthy snacks, aiming to address the detrimental environmental and economic impacts of discarding such waste. Through direct ink writing, edible constructs containing orange peel waste were successfully created. Biochemical analysis and cell viability tests revealed that the nutritional and antioxidant content of the orange peel waste was not affected during the manufacturing process. This study highlights the potential of revalorizing food waste and achieving more sustainable and circular food manufacturing and consumption through 3D printing.

Spatiotemporal, physiological and transcriptomic dynamics of wild jujube seedlings under saline conditions

Aobing He, Zhibo Ma, Yunfei Li, Chen Huang, Jean Wan Hong Yong, Jian Huang

Summary: Soil salinity is a major constraint for jujube production in China. Wild jujube is used as the rootstock to overcome saline conditions. This study investigated the adaptive mechanism of wild jujube under salt stress through spatiotemporal, physiological, and transcriptomic analyses. The results showed that salt treatments had dose and duration effects on biomass, photosynthesis, and ion accumulation. Differential expression genes were identified and functional enrichment analysis revealed altered physiological activities. Certain genes, including nitrogen and potassium transporters and WRKY/MYB/bHLH transcription factors, were identified as hub genes responding to salt stress.
