Regulation of IL-17 production in human lymphocytes

The discovery of a new lineage of helper T cells that selectively produces interleukin (IL)-17 has provided exciting new insights into immunoregulation, host defense and the pathogenesis of autoimmune diseases. Although the factors that promote murine Th17 differentiation have been intensively examined, there has been much less information on the regulation of this cytokine in human T cells. IL-17 is readily produced by human memory T cells, which we now know exhibit distinct patterns of chemokine receptor expression and may differentiate in response to selective pathogens. Recently it has been shown that IL-1, IL-6 and IL-23 are important in driving human Th17 differentiation. However, TGFP-1 which is important for the differentiation of murine Th17 cells and inducible regulatory T cells (iTregs), is reportedly not required and even inhibits for human Th17 differentiation. In addition, human Th17 cells also produce other proinflammatory cytokines. Further characterization of the transcription regulation of human IL-17 expression, and the epigenetic regulation of human Il17 locus should improve our understanding the lineage commitment of human Th17 cells. Targeting the production and action of this cytokine is also likely to be beneficial therapeutically for autoinflammatory and autoimmune diseases. Published by Elsevier Ltd.

Vimentin Suppresses Inflammation and Tumorigenesis in the Mouse Intestine

Summary: Vimentin plays an important role in intestinal diseases, particularly in the injury and repair processes of colonic epithelial cells. Vim(-/-) mice showed more severe colitis and higher production of reactive oxygen and nitrogen species than wild-type mice when exposed to DSS. CD11b(+) macrophages were identified as the main cellular source of reactive oxygen species production. Additionally, Vim(-/-) mice did not spontaneously develop colitis-associated cancer model upon DSS treatment, but exhibited increased tumor numbers and size in the distal colon in the azoxymethane/DSS model.

Plasma Proteomics Unveil Novel Immune Signatures and Biomarkers upon SARS-CoV-2 Infection

Summary: Using a proximity extension assay, the study quantified 1387 proteins in plasma samples from 28 Finnish patients with COVID-19 and their controls to determine the protein profile changes caused by SARS-CoV-2 infection. The findings revealed altered immune signatures in patients with comorbidities and the identification of novel elevated proteins, anti-microbial activity proteins, and extracellular matrix remodeling proteins. Additionally, downregulation of RNF41 was observed in patients compared to healthy controls. These plasma protein changes provide insights into SARS-CoV-2 infection and potential biomarkers for diagnosis, prognosis, and therapeutic targets.

This study investigated the potential involvement of macrophage-derived exosomes in the pathogenesis of cryptococcosis. The study analyzed the changes of microRNAs in macrophage exosomes and explored the effects of highly expressed exo-miRNAs on human endothelial cell permeability, ROS accumulation, and pyroptosis in bronchial epithelioid cells. Inflammatory marker levels in children with tobacco smoke exposure

Suppression of DNMT2/3 by proinflammatory cytokines inhibits CtBP1/ 2-dependent genes to promote the occurrence of atrophic nonunion

Summary: Failure of bone healing after fracture often results in nonunion, but the underlying mechanism of nonunion pathogenesis is poorly understood. This study reveals that the inflammatory microenvironment in atrophic nonunion inhibits the expression of DNA methyltransferases, resulting in overexpression of CtBP1/2. The increased CtBP1/2 acts as transcriptional corepressors that suppress the expression of six genes involved in bone healing. The disruption of this signaling may represent a new therapeutic strategy to prevent nonunion incidence after bone fracture.
