The molecular mechanisms underlying myotube migration are not well understood, but studies in Drosophila have identified some molecular signals involved in this process. Summary: Dot1l is a histone methyltransferase responsible for H3K79 methylation, but it also participates in gene expression silencing. In this study, we found that Dot1l represses endogenous retroviruses and activates the 2-cell-like state in embryonic stem cells (ESCs). Mechanistically, Dot1l interacts with Npm1 to suppress MERVL expression by regulating protein level and deposition of histone H1. Our findings reveal a non-canonical transcriptional repressive role of Dot1l in ESCs.

Furthermore, the study shows that Hikeshi-dependent hyperthermia therapy can promote remyelination.

Summary: Repeated absence of useable embryos is a challenging issue in infertility patients, with embryonic developmental arrest being commonly observed but genetically unknown. In this study, the arrested embryonic development in a patient was thoroughly documented through photo-taking, and a homozygous PADI6(R132C) variant was identified through whole-exome sequencing. Analysis of conservativeness and structural impact of the variant provided further evidence for the crucial role of homozygous PADI6(R132C) variant in female infertility. This study contributes to a deeper understanding of the molecular genetic basis of female infertility.

Summary: Holometabolous insects show significant changes in gene expression during metamorphosis, resembling the embryonic stage rather than the larval stage. The transition from pupa to imago shows similar gene expression patterns as the transition from embryo to larva. Genes associated with metabolism and development undergo changes in expression levels during the larval stage and revert to an embryonic-like state during metamorphosis.
