Quantification of p38αMAP kinase: A prognostic marker in HNSCC with respect to radiation therapy

Background: Translation of early findings from basic research is aimed to benefit cancer therapeutics. We report the p38 alpha level in serum of head and neck squamous cell carcinoma (HNSCC) patients indicating it as a prognostic marker and established its correlation with radiation therapy (RT). Methods: The case-controlled study was performed on 120 HNSCC patients from whom 81 patients and 45 controls were statistically analyzed. The p38 alpha estimation was done at pre-RT, during-RT and post-RT using a real time Surface Plasmon Resonance (SPR) technology, ELEA and western blot. Results: HNSCC patients showed threefold increase in p38 alpha level when compared to control (p value <0.0001). The estimated concentration of p38 alpha in a temporal manner, before-RT, during-RT and post-RT was 0.61 ng/mu l (95%CI: 0.53-0.69), 035 ng/mu l (95%CI: 0.31-038) and 0.30 ng/mu l (95%CI: 0.26-033), respectively. Among the 81 cases, 70 patients (86.42%) showed a declined p38 alpha in response to RT as evaluated by SPR and were responding clinically (clinical tumor regression). Conclusions: This study showed elevated p38 alpha level at cancer diagnosis and a statistically significant decline during-RT and post-RT periods. Hence, it can emerge as a prognostic marker supporting the candidature of p38 alpha as a suitable serum marker in HNSCC. (C) 2011 Elsevier B.V. All rights reserved.

Role of Midkine in Cancer Drug Resistance: Regulators of Its Expression and Its Molecular Targeting

Summary: Molecules involved in drug resistance can be targeted for better therapeutic efficacies. Research on midkine (MDK) has shown a positive correlation between disease progression and MDK expression in most cancers, indicating its association with multi-drug resistance. MDK, a secretory cytokine found in blood, can be used as a potent biomarker for non-invasive detection of drug resistance in cancer and can be targeted for therapy. This article summarizes the current knowledge on the involvement of MDK in drug resistance and its transcriptional regulation, highlighting its potential as a therapeutic target for cancer.

Summary: Traditional cancer treatment faces obstacles such as drug resistance, toxicity, and financial constraints. Phytochemicals that regulate molecular mechanisms are gaining interest as alternative medicine. This study used network pharmacology to explore the regulatory mechanisms of 6-shogaol in cervical cancer treatment. Various databases were screened to identify target genes of 6-shogaol and disease-related targets. Protein-protein interactions and topology analysis identified hub genes. Gene ontology and pathway enrichment analysis were conducted, and molecular docking and survival analysis revealed potential target genes for treatment. The study provides scientific evidence supporting the use of 6-shogaol as a treatment for cervical cancer.

However, the high operational costs and limited production volume are major limitations in this process. Recent studies have investigated the use of nanocatalysts in biofuel production to overcome these challenges. This review discusses the application of nanocatalysts in different stages of biofuel production and their advantages and disadvantages.

Summary: In recent years, there has been a global threat to human health and environment due to the development of antimicrobial resistance (AMR) in bacterial pathogens. In this study, silver nanoparticles derived from Lactobacillus rhamnosus (AgNPs-LR) were tested against three Gram-negative bacteria to determine their effects on biofilm formation and virulence factors controlled by quorum sensing (QS). The results demonstrated that AgNPs-LR effectively inhibited the development of biofilms and QS-mediated virulent traits of these bacteria.

Secondary Metabolite Profiling, Antioxidant, Antidiabetic and Neuroprotective Activity of Cestrum nocturnum (Night Scented-Jasmine): Use of In Vitro and In Silico Approach in Determining the Potential Bioactive Compound

Summary: This study aims to investigate the therapeutic potential of C. nocturnum leaf extracts against diabetes and neurological disorders through inhibitory activities on alpha-amylase and acetylcholinesterase (AChE). Computational molecular docking studies support the inhibitory potential of C. nocturnum leaves-derived secondary metabolites on alpha-amylase and AChE. The methanolic fraction of the leaf extract showed strong antioxidant activity and inhibited alpha-amylase and AChE in a non-competitive and competitive manner, respectively. In silico analysis revealed high-affinity binding of compounds in the methanolic extract to the active sites of alpha-amylase and AChE.

Summary: Rhizomes of Curculigo orchioides Gaerten. (Hypoxidaceae) were extracted with hydroalcoholic solvent and separated into alkaloidal and non-alkaloidal fractions. Both fractions showed dose-dependent anti-inflammatory effects, with the alkaloidal fraction being more effective in HRBC method and the non-alkaloidal fraction being more effective in PD method. The study also identified various phytochemicals in C. orchioides that may contribute to its anti-inflammatory action.

Both genetic and non-genetic factors contribute to the co-occurrence of CHD and NDD. Further research should focus on identifying common molecular mechanisms underlying this co-occurrence and promoting the research and treatment of developmental disorders related to the cardiovascular and central nervous systems.

Accurate identification of HLA-B*15:02 allele by two-dimensional polymerase chain reaction

Summary: This study successfully established a 2D-PCR method for identifying HLA-B*15:02 through a two-tube reaction. This method can distinguish HLA-B*15:02 from 16 highly homologous HLA-B*15 alleles. Among 1830 samples from the clinical general population, 3 HLA-B*15:02 homozygotes and 84 HLA-B*15:02 heterozygotes were detected.
