Summary: This study investigated the role of GPR146 in pulmonary arterial hypertension (PAH) and found that GPR146 is highly expressed in the pulmonary vessels. It was discovered that GPR146 promotes pulmonary vascular remodeling by promoting the proliferation of pulmonary artery smooth muscle cells (PASMCs) through the upregulation of 5-lipoxygenase (5-LO) expression. This finding suggests that GPR146 may serve as a possible target for the prevention and treatment of PAH.

Summary: This study found that the p38MAPK/DUSP1 negative feedback loop is impaired in pulmonary arterial hypertension (PAH), leading to excessive activation of p38MAPK and hyperplasia of PA-SMCs. The IP receptor agonist MRE-269 increases DUSP1 expression and inhibits p38MAPK-mediated PA-SMC proliferation. Further elucidation of the mechanism underlying reduced DUSP1 expression would be informative for PAH treatment.

Summary: The functional diversity of K+ channels in the pulmonary vasculature is attributed to various factors including gene expression, alternative RNA splicing, post-transcriptional modifications, and the presence of different subunits. These channels play a crucial role in regulating membrane potential and controlling the dilation or contraction of pulmonary arteries. Impaired K+ channel activity is a key factor in the development of pulmonary hypertension, which can result from mutation, downregulation of expression, or decreased channel current due to various vasoactive factors or drug exposure. Restoring K+ channel function is a potential therapeutic strategy for pulmonary hypertension.

Summary: This study investigated the role of nPKC in mediating PASMCs proliferation in PH. The findings revealed that hypoxia promoted PASMCs proliferation, with the most significant effect observed after 72 hours of treatment. The upregulation of PKC8 and PKC? expression under hypoxia led to PASMCs proliferation via the upregulation of AKT and ERK phosphorylation. These results suggest potential novel targets for molecular therapy of pulmonary hypertension.
