Proximal tubular Bmal1 protects against chronic kidney injury and renal fibrosis by maintaining of cellular metabolic homeostasis

Summary: Recent studies have shown that the core clock gene Bmal1 plays a significant role in renal physiological functions. This study found that deletion of Bmal1 in the proximal tubule exacerbated chronic kidney disease (CKD) and renal fibrosis. Mechanistically, Bmal1 regulates fatty acid oxidation and glutathione metabolism, and activates the transcription of CBS to stimulate glutathione biosynthesis. These findings provide a potential target for treating CKD.

Circadian control of ConA-induced acute liver injury and inflammatory response via Bmal1 regulation of Junb

Summary: This study reveals a critical role of the Bmal1-Junb-AKT/ERK axis in the circadian control of ConA-induced hepatitis, providing new insights into the prevention and treatment of immune-mediated hepatitis, including autoimmune hepatitis (AIH). The findings have scientific implications as they enhance our understanding of the circadian regulation of immune responses in liver diseases.

Pemetrexed ameliorates Con A-induced hepatic injury by restricting M1 macrophage activation

Summary: This study demonstrates that Pemetrexed (PEM) effectively alleviates immune-mediated hepatic injuries by inhibiting M1 macrophage activation through the NF-kappa B signaling pathways. It significantly improves mouse survival rates and reduces serum transaminase levels. Additionally, PEM also mitigates oxidative stress, reduces histopathological liver damage, and decreases hepatocyte apoptosis. Furthermore, PEM decreases the expression of proinflammatory cytokines and genes associated with M1 macrophages in the liver.
