Medicinal Organometallic Chemistry: Designing Metal Arene Complexes as Anticancer Agents

The field of medicinal inorganic chemistry is rapidly advancing. In particular organometallic complexes have much potential as therapeutic and diagnostic agents. The carbon-bound and other ligands allow the thermodynamic and kinetic reactivity of the metal ion to be controlled and also provide a scaffold for functionalization. The establishment of structure-activity relationships and elucidation of the speciation of complexes under conditions relevant to drug testing and formulation are crucial for the further development of promising medicinal applications of organometallic complexes. Specific examples involving the design of ruthenium and osmium arene complexes as anticancer agents are discussed.

Summary: Research suggests that the structural stability of bevacizumab with CPP is not affected by temperature, but physical instability and aggregation occur after five weeks. The addition of the polypeptide leads to increased aggregation, but the complexing of bevacizumab with CPP has no effect on its biological stability or degradation.

Location-Dependent Lanthanide Selectivity Engineered into Structurally Characterized Designed Coiled Coils

Summary: This report highlights the unprecedented phenomenon of location-dependent and size-selective binding to lanthanide sites within miniature protein coiled coil scaffolds. The engineered binding sites show unusual selectivity for moderately large Ln(3+) ions, and it is demonstrated that this selectivity can be programmed into the sequence and manipulated by shifting the binding site. Experimental data, including X-ray crystal structures and CD studies, provide insights into the optimal radius of binding sites within the coiled coil scaffold, contributing to a better understanding of location-dependent metal selectivity in protein structures.

Summary: This study discovered a simple and effective method to induce titan cells in C. gattii under in vitro conditions, explored the titanisation ability in immunocompetent individuals, and characterized the differences among strains. The study also found that genes associated with cell cycle progression played a crucial role in titanisation, and identified exogenous p-Aminobenzoic acid as a key inducer of titanisation.

By designing α-helical coiled coil peptides as scaffolds, control over the coordination environment of lanthanide complexes can be achieved, contributing to the expansion of native lanthanide biochemistry.
