Summary: The study found that DMF can inhibit nitric oxide and proinflammatory cytokine production in microglia, induce microglial switching to the M2 state, increase the expression levels of autophagy markers such as LC3 and ATG7, and promote the formation of autophagosomes. These results suggest that DMF induces autophagy in microglia and its anti-inflammatory effects are partially mediated through an autophagy-dependent pathway.

Summary: The study found that HMGB1 plays a critical role in mediating NOD2-induced autophagy in microglia. Cytoplasmic accumulation of HMGB1 activates a cell-autonomous pro-autophagic pathway, while extracellular secretion of HMGB1 activates a cell-autonomous pro-inflammatory pathway.

Summary: This study revealed dysfunction of the autophagic process in chronic migraine, and suggested that activated autophagy may have a preventive effect on migraine chronification. P2X7R contributes to central sensitization through mediating autophagy regulation, making it a potential therapeutic target for chronic migraine.

Summary: The study found that Vesatolimod, a toll-like receptor 7 agonist, has significant therapeutic effects on Experimental autoimmune encephalomyelitis (EAE) mice. It reduces clinical symptoms, inflammatory cytokine levels, demyelination, and immune cell infiltration in the spinal cords. These findings suggest that Vesatolimod could be a promising treatment for multiple sclerosis.

Summary: Disturbance of macrophage-associated lipid metabolism plays a key role in atherosclerosis. The crosstalk between autophagy deficiency and inflammation response in foam cells (FCs) through epigenetic regulation is poorly understood. In this study, it was observed that oxidized low-density lipoprotein (ox-LDL) led to macrophage autophagy deficiency and abnormal crosstalk between autophagy and inflammation. The dysfunctional transcription factor EB (TFEB)-P300-bromodomain-containing protein 4 (BRD4) axis mediated aberrant lipid metabolism. ox-LDL activated BRD4-dependent super-enhancer associated with liquid-liquid phase separation (LLPS) on the regulatory regions of inflammatory genes. Curcumin (Cur) restored FCs autophagy by promoting TFEB nuclear translocation, optimizing lipid catabolism, and reducing inflammation. The anti-atherogenesis effect of Cur was inhibited by macrophage-specific Brd4 overexpression or Tfeb knock-out in Apoe knock-out mice via bone marrow transplantation. These findings establish a novel TFEB-P300-BRD4 axis and highlight the epigenetic regulatory mechanisms by which Cur regulates autophagy, inhibits inflammation, and decreases lipid content.

Summary: KOS can protect microglia from over-activated inflammation by inducing autophagy and inhibiting the release of inflammatory cytokines. Additionally, KOS can regulate the autophagy pathway and inhibit the inflammatory response of over-activated microglia by reducing the expression of proteins related to the AMPK/ULK1 pathways.

Summary: Alzheimer's disease is a debilitating neurological disorder with no current cure. Pathological features such as amyloid plaques and tau tangles are characteristic of AD, but targeting these has not shown desired efficacy in patients. Autophagy, inflammation, and microglia play significant roles in AD pathogenesis and may provide potential therapeutic targets.
