The epigenetic role of HTR1A antagonist in facilitating GnRH expression for pubertal initiation control

Shasha Zhou, Yihang Shen, Shaolian Zang, Xiaoqin Yin, Pin Li

Summary: In this study, HTR1A antagonists were found to enhance GnRH transcription by promoting PRC1 degradation and H2AK119ub loss through reduced CBX4 expression via suppression of the PI3K/Akt and MAPK/ERK pathways in GT1-7 cells, potentially providing an epigenetic mechanism of action of HTR1A on GnRH gene expression for mammalian puberty onset.

Transcriptome-scale spatial gene expression in rat arcuate nucleus during puberty

Shasha Zhou, Shaolian Zang, Yanping Hu, Yifen Shen, Hua Li, Wenlian Chen, Pin Li, Yihang Shen

Summary: This study investigated the gene expression patterns in different pubertal stages in the hypothalamus and identified a subset of kisspeptin neurons using a specific membrane surface biomarker. It was also found that different cell types in the hypothalamus have distinct functions in neurogenesis, hormone secretion, and estradiol response, contributing to the complex regulatory network of the hypothalamic-pituitary-gonadal axis.

Summary: The study revealed that changes in DNA hydroxymethylation and DNA methylation were associated with gene expression patterns during puberty in the arcuate nucleus, with SINEs playing a regulatory role in gene expression.

Genomic distribution of signal transducer and activator of transcription (STAT) family in colorectal cancer

Yanping Hu, Yifen Shen, Yang Zhao, Ying Tang, Chao Liu, Yongchun Gu, Tao Yang, Yihang Shen

Summary: In this study, ChIP-seq was used to investigate the genome-wide distribution of activated STAT proteins in colorectal cancer cells. It was found that the homodimers of STAT proteins predominantly occupied genomic DNA and STAT3 accounted for the largest proportion. The binding motif targeted by different STAT homodimers suggested distinct biological functions. The study also revealed NR5A2 as a specific co-activator of STAT3 and its role in CRC. These findings provide new insights into the design of STAT inhibitors for treating CRC and other diseases.

Summary: The study found that CD8 + CD279+ cells are an independent prognostic factor for DFS in CRC patients, with high levels of CD8 + CD279+ cells associated with better DFS.
