The chemokine SDF-1a and its receptor CXCR4b guide germ cell migration in zebrafish by activating downstream signaling events. Boldajipour et al. (2008) now report that a second SDF-1a receptor, CXCR7, is also required for guided migration but does not function as a signaling receptor, and instead sequesters SDF-1a. These results highlight the importance of ligand clearance during guided cell migration.

Knock-in tagging in zebrafish facilitated by insertion into non-coding regions

Summary: A simple approach for tagging proteins in zebrafish on their N or C termini with fluorescent proteins by inserting PCR-generated donor amplicons into non-coding regions of the corresponding genes has been developed, enabling the generation of endogenously tagged alleles for crucial genes in epithelial biology and organ development. This approach facilitates the generation of knock-in lines in zebrafish, paving the way for accurate quantitative imaging studies.

The pattern of nodal morphogen signaling is shaped by co-receptor expression

Summary: Embryos communicate instructions to cells through morphogens, which form gradients by diffusing from a source. However, natural patterning systems involve co-factors and signaling feedback, suggesting additional mechanisms are needed for signaling patterns. For the mesendoderm inducer Nodal in zebrafish embryos, the range of Nodal ligands is regulated by the EGF-CFC co-receptor Oep to control the spread and sensitivity of signaling, as confirmed by computational modeling and experimental observations.
