Preclinical Validation of Electrochemotherapy as an Effective Treatment for Brain Tumors

Electrochemotherapy represents a strategy to enhance chemotherapeutic drug uptake by delivering electrical pulses which exceed the dielectric strength of the cell membrane, causing transient formation of structures that enhance permeabilization. Here we show that brain tumors in a rat model can be eliminated by electrochemotherapy with a novel electrode device developed for use in the brain. By using this method, the cytotoxicity of bleomycin can be augmented more than 300-fold because of increased permeabilization and more direct passage of drug to the cytosol, enabling highly efficient local tumor treatment. Bleomycin was injected intracranially into male rats inoculated with rat glia-derived tumor cells 2 weeks before the application of the electrical field (32 pulses, 100 V, 0.1 ms, and 1 Hz). In this model, where presence of tumor was confirmed by magnetic resonance imaging (MRI) before treatment, we found that 9 of 13 rats (69%) receiving electrochemotherapy displayed a complete elimination of tumor, in contrast to control rats treated with bleomycin only, pulses only, or untreated where tumor progression occurred in each case. Necrosis induced by electrochemotherapy was restricted to the treated area, which MRI and histology showed to contain a fluid-filled cavity. In a long-range survival study, treatment side effects seemed to be minimal, with normal rat behavior observed after electrochemotherapy. Our findings suggest that electrochemotherapy may offer a safe and effective new tool to treat primary brain tumors and brain metastases. Cancer Res; 71(11); 3753-62. (C)2011 AACR.

Quality-of-life outcomes following topical melatonin application against acute radiation dermatitis in patients with early breast cancer: A double-blind, randomized, placebo-controlled trial

Summary: This study aimed to investigate the impact of topical melatonin during radiation therapy on the quality of life in primary breast cancer patients. A total of 65 patients were included in the study, with 26 in the melatonin group and 22 in the placebo group. The results showed that while there was no difference in breast symptom scores on the last day of radiation, the melatonin group had significantly lower scores over the entire duration of the trial.

Processes of Care and Associated Factors in Patients With Stroke by Immigration Status

Summary: This study investigated the differences in acute stroke care between immigrants and Danish-born residents, and found that immigrants were less likely to receive early stroke care and be admitted to stroke unit, as well as received fewer individualized stroke care processes. These disparities were primarily influenced by the socioeconomic status and cohabitation of immigrants.

Decomposition-based framework for tumor classification and prediction of treatment response from longitudinal MRI

Summary: A new prediction framework was proposed in this study to utilize temporal dynamics of tissue features from repeated measurements. The framework was able to classify tumor types and predict the response to radiotherapy using MRI data analysis. The results showed accurate classification of pancreatic tumor types and prediction of tumor volume change. The classification of brain metastases also demonstrated some level of accuracy.

Summary: The objective of this study was to define QA tests for oMRgRT systems and to identify important medical physics aspects for installation and commissioning. Twenty-four potentially important QA tests were identified, along with eleven tasks and aspects related to construction, workflow development, and training. A program for QA and commissioning of oMRgRT systems was developed to support safe handling of such systems.

Cortical hemodynamic response during cognitive Stroop test in acute stroke patients assessed by fNIRS

Summary: This study explored the feasibility of fNIRS as a biomarker to assess executive function in AIS patients. The results showed that AIS patients performed worse than the healthy controls in the SCWT. However, both groups exhibited an inverse hemodynamic response in the prefrontal cortex during the test.

An Explorative Investigation of the Effect of Naturalistic Light on Agitation-Associated Behavior in Nursing Home Residents With Dementia: A Pilot Study

Summary: Electrochemotherapy is an effective treatment for cutaneous metastases of various cancers, including breast cancer. This study analyzed breast cancer patients with different receptor statuses (estrogen receptor and HER2 receptor) using the INSPECT database. The study found that ECT treatment was equally effective among different groups, and treating multiple small lesions at the time of occurrence was more effective than treating larger, long-lasting armor-like lesions.

Quantification of cross-vendor variation in ADC measurements in vendor-specific prostate MRI-protocols

Summary: The purpose of this study was to quantify the variability of ADC and investigate if there were significant differences in ADC between MRI systems and sequences. A phantom model with fixed ADC values was tested in six MRI systems using different sequences. Statistical analysis revealed significant differences in ADC measurements between vendors, while the variation of ADC between MRI systems and sequences was deemed clinically irrelevant.

Impact of abdominal compression on intra-fractional motion and delivered dose in magnetic resonance image-guided adaptive radiation ablation of adrenal gland metastases

Summary: This study investigated the impact of abdominal compression on motion and the delivered dose during non-gated, MRI-guided radiation ablation of adrenal gland metastases. The results showed that the use of abdominal compression significantly reduced the target position error and prevented the degradation of target coverage. Most patients maintained acceptable target coverage and dose to intra-luminal organs during motion.

Summary: This study aimed to evaluate the variation in ADC measurements and calculations among multiple centers and provide recommendations to minimize it. The results showed that calculation methods had a bigger impact on ADC reproducibility compared to delineation methods. It is suggested to consider these factors when conducting multi-center studies.

A Systematic Review of the Circadian Rhythm of Bone Markers in Blood

Summary: This review provides an overview of the circadian variation of bone markers and the factors that influence this rhythmicity. Many bone markers show a marked circadian variation with nighttime or early morning peaks. Factors such as fasting can affect the circadian variation of these markers. Understanding and considering these circadian variations and influencing factors are important for accurate measurement of bone markers.
