Summary: HIF-1 alpha is a protein widely distributed in human cells that can mediate hypoxia signals and regulate compensatory responses to hypoxia. In recent years, researchers have discovered HIF-1 alpha inhibitors, which may become a new class of drugs for treating diseases such as tumors, leukemia, diabetes, and ischemic diseases.

Summary: Uncontrolled growth of solid tumors can lead to hypoxic conditions and activation of HIF-1 alpha, promoting metabolism, proliferation, and metastasis of tumor cells. Inhibiting the transcriptional activity of HIF-1 could be a potential strategy for cancer therapy. Nbs selected through phage display showed significant inhibitory effects on the transcriptional activity of HIF-1, confirming their potential in cancer treatment.

Summary: High-dose hypofractionated radiation stimulates anti-tumor immune response but also increases immune suppression mediated by HIF alpha. Inhibition of HIF-1 alpha can restore radiation-induced anti-tumor immunity. Combination of HIF-1 alpha inhibitors with immune checkpoint blocking antibodies may enhance overall anti-tumor immune profile.

Summary: This study investigates the effect of oxygen and the oxygen stress regulator HIF-1 alpha on cholesterol-dependent NET formation. The researchers found that the formation of NETs was significantly induced by M beta CD under both normoxic and hypoxic conditions in murine neutrophils and human neutrophils. Similar observations were made in human neutrophils stimulated with M beta CD or statins.

Summary: This study introduces a new strategy of building a ROS FET to address the challenge of tumor hypoxia, achieving effective tumor ablation both in vitro and in vivo. The mechanism of changing tumor hypoxia is revealed, creating a conceptually novel method for maximizing ROS damages for cancer treatment.

Summary: This study discovered a natural compound, steppogenin, that inhibits both HIF-1 alpha and DLL4, leading to the suppression of angiogenesis and tumor growth. The experimental results showed that steppogenin exhibited significant anti-angiogenic and anti-tumor activities in cell and animal experiments. This research provides a potential candidate compound for developing novel therapies for angiogenic diseases, including solid tumors.

Summary: The study revealed that the overexpression of HECTD2 and HIF-1 alpha was associated with poorer overall survival in RCC patients, and their overexpression increased RCC cell proliferation and migration while reducing apoptosis. Conversely, knocking down HECTD2 or HIF-1 alpha impeded the malignant phenotypes of RCC cells.

Summary: The Pringle maneuver (PM) used during hepatocellular carcinoma (HCC) resection can lead to tumor metastasis due to ischemia and hypoxia. This study discovered that high expression of suppressor of cytokine signaling 5 (SOCS5) significantly affected the prognosis of HCC patients undergoing long-time PM. The study also showed that SOCS5 promotes the expression of hypoxia-inducible factor 1 subunit alpha (HIF-1 alpha) protein, which is associated with higher hypoxia scores. Knockdown of SOCS5 inhibited HCC cell invasion and migration through the PI3K/Akt/mTOR/HIF-1 alpha signaling axis. This research highlights the potential of a SOCS5-specific inhibitor to control PM-induced tumor micrometastases during HCC resection.

Summary: This study retrospectively analyzed the prognostic factors and treatment outcomes of primary malignant mediastinal germ cell tumors (GCTs) in Taiwanese children. The results showed that surgical treatment was associated with better outcomes, while advanced disease stage and choriocarcinoma histology subtype were associated with poorer outcomes. Additionally, there was no difference in treatment outcomes between cisplatin-based and carboplatin-based chemotherapy.
