Attenuation of TNF production and experimentally induced inflammation by PDE4 inhibitor rolipram is mediated by MAPK phosphatase-1

BACKGROUND AND PURPOSE 3',5'-Cyclic nucleotide PDE4 is expressed in several inflammatory and immune cells, and PDE4 catalyses the hydrolysis of cAMP to 5'AMP, down-regulating cAMP signalling in cells. MAPK phosphatase-1 (MKP-1) is an endogenous p38 MAPK signalling suppressor and limits inflammatory gene expression and inflammation. In the present study, we investigated the effect of a PDE4 inhibitor rolipram on MKP-1 expression and whether MKP-1 is involved in the anti-inflammatory effects of rolipram. EXPERIMENTAL APPROACH The effect of rolipram on TNF production was investigated in J774 mouse macrophage cell line and in primary mouse peritoneal macrophages (PM) from wild-type (WT) and MKP-1(-/-) mice. We also investigated the effect of rolipram on carrageenan-induced paw inflammation in WT and MKP-1(-/-) mice. KEY RESULTS MKP-1 expression was enhanced by rolipram, by a non-selective PDE inhibitor IBMX and by a cAMP analogue 8-Br-cAMP in J774 cells and in PM. Enhanced MKP-1 mRNA expression by rolipram was reversed by a PKA inhibitor. Rolipram, IBMX and 8-Br-cAMP also inhibited TNF production in activated macrophages. Accordingly, rolipram inhibited TNF production in PMs from WT mice but, interestingly, not in PMs from MKP-1(-/-) mice. Furthermore, rolipram attenuated carrageenan-induced paw inflammation in WT but not in MKP-1(-/-) mice. CONCLUSIONS AND IMPLICATIONS PDE4 inhibitor rolipram was found to enhance the expression of MKP-1, and MKP-1 mediated, at least partly, the anti-inflammatory effects of PDE4 inhibition. The results suggest that compounds that enhance MKP-1 expression and/or MKP-1 activity hold potential as novel anti-inflammatory drugs.

Summary: This study aimed to describe the profile of severe asthma patients eligible for both anti-IgE and anti-IL5/5R and compare the effectiveness of these two treatments in real-life settings. The results showed that anti-IL5/5R was superior to anti-IgE in reducing asthma exacerbations and long-term oral corticosteroid use, while there was some evidence suggesting a decrease in asthma-related hospitalizations with anti-IL5/5R treatment.

Summary: This study aimed to investigate the influence of abdominal fat content, as judged by waist:height ratio, on blood pressure and hemodynamic profile. The results showed that individuals with higher waist:height ratios had higher blood pressure, systemic vascular resistance, and arterial stiffness. These findings suggest that obesity-related hypertension has similar characteristics to primary hypertension.

Summary: This study found that human osteoarthritic chondrocytes express 19 different TRP genes, with significant expression of TRPM8. Four distinct trajectories of severe asthma were identified, illustrating different patterns of progression of asthma severity. This study may contribute to the development of better preventive management strategies for severe asthma.

The regulation of TRPA1 expression and function by Th1 and Th2-type inflammation in human A549 lung epithelial cells

Summary: This study investigated the effects of Th1 and Th2-type inflammation on TRPA1 expression and function. The results showed that TRPA1 expression and function were enhanced under inflammatory conditions, with IFN-γ increasing TRPA1 expression and IL-4 and IL-13 suppressing it in a JAK-STAT6 dependent manner. These findings suggest that the paradigm of Th1 and Th2 inflammation is a major determinant of TRPA1 expression and function in (lung) disease and should be considered in pharmacotherapy.

Type 2 Low Biomarker Stability and Exacerbations in Severe Uncontrolled Asthma

Summary: We investigated the stability of T2 low status and exacerbation rates in T2 low and non-T2 low asthma using clinical retrospective data. Among patients with severe uncontrolled asthma, 18% were identified as T2 low(150) and 39% as T2 low(300). Biomarker stability was observed in 55% and 72% of patients in these groups. Exacerbation rates were higher in the T2 low group compared to the non-T2 low group. BEC and FeNO are useful biomarkers in identifying and monitoring T2 low asthma.

Ultra-low-dose CBCT: new cornerstone of paranasal sinus imaging
