Quantification of Histone Modifications by Parallel-Reaction Monitoring: A Method Validation

Abnormal epigenetic reprogramming is one of the major causes leading to irregular gene expression and regulatory pathway perturbations, in the cells, resulting in unhealthy cell development or diseases. Accurate measurements of these changes of epigenetic modifications, especially the complex histone modifications, are very important, and the methods for these measurements are not trivial. By following our previous introduction of PRM to targeting histone modifications (Tang, H.; Fang, H.; Yin, E.; Brasier, A. R.; Sowers, L. C.; Zhang, K. Multiplexed parallel reaction monitoring targeting histone modifications on the QExactive mass spectrometer. Anal. Chem. 2014, 86 (11), 5526-34), herein we validated this method by varying the protein/trypsin ratios via serial dilutions. Our data demonstrated that PRM with SILAC histones as the internal standards allowed reproducible measurements of histone H3/H4 acetylation and methylation in the samples whose histone contents differ at least one-order of magnitude. The method was further validated by histones isolated from histone H3 K36 trimethyltransferase SETD2 knockout mouse embryonic fibroblasts (MEF) cells. Furthermore, histone acetylation and methylation in human neural stem cells (hNSC) treated with ascorbic acid phosphate (AAP) were measured by this method, revealing that H3 K36 trimethylation was significantly down-regulated by 6 days of treatment with vitamin C.

Inhibition of the Akt/PKB Kinase Increases Nav1.6-Mediated Currents and Neuronal Excitability in CA1 Hippocampal Pyramidal Neurons

Summary: In this study, it was found that Akt inhibition enhances peak transient Na+ current density through Na(v)1.6 channels and increases action potential firing in CA1 pyramidal neurons, accompanied by a decrease in action potential current threshold. Complementary confocal analysis revealed that Akt inhibition is associated with the lengthening of Na(v)1.6 fluorescent intensity along the axonal initial segment, providing a mechanism for augmented neuronal excitability.

Efficacy of Constructing Digital Hybrid Skull-Dentition Images Using an Intraoral Scanner and Cone-Beam Computed Tomography

Summary: This study aimed to evaluate the potential for improving dentition imaging with CBCT scans using an intraoral scanner instead of a plaster model. Summary: This study investigated the relationship between skeletal and soft tissue changes after incisor retraction in patients with Angle Class I bimaxillary dentoalveolar protrusion. The results showed a significant correlation between skeletal point A and soft tissue point A on the anteroposterior axis. However, changes in incisor tip, apex, and inclination did not significantly affect the position of skeletal points A and B. It is important to consider both hard and soft tissue analysis in treatment planning for malocclusion.

Predictors of bullying, depression, and suicide attempts among youth: The intersection of race/ethnicity by gender identity

Summary: Using multivariate logistic regression, this study analyzed data from the 2015 Healthy Kids Colorado Survey to explore the relationship between the intersection of race/ethnicity and gender identity with mental and behavioral health-related risks. The findings suggest that youth with multiple marginalized identities, particularly those who identify as both Bi/Multi-racial or Latino and as transgender/non-binary, have higher levels of depression and suicidal ideation compared to cisgender White youth. Intersectional approaches are important in designing risk prevention and response interventions for racial/ethnic minority youth who are also transgender/non-binary.

Effectiveness of cone-beam computed tomography-generated cephalograms using artificial intelligence cephalometric analysis

Summary: Lateral cephalograms and related analysis are representative methods for orthodontic treatment, but inaccuracies may be produced when quantitative evaluation is required. Cone-beam computed tomography (CBCT) provides high-resolution three-dimensional images at a relatively low dose and cost, and using artificial intelligence analysis programs for lateral cephalogram generation from CBCT has proven to be efficient and consistent for orthodontic diagnosis and planning.

Fracture and Fatigue of Dental Implants Fixtures and Abutments with a Novel Internal Connection Design: An In Vitro Pilot Study Comparing Three Different Dental Implant Systems

Summary: The aim of this study was to compare the mechanical behaviors of three dental implant fixtures with different abutment connection designs. The results of the static compression to fracture tests and fatigue tests showed that all three implants had suitable mechanical properties for intraoral use. Fracture patterns may vary due to differences in the fixture design or abutment connection.

Parent-Worker Relationships in Home Visiting: Intersections of Parental Gender, Ethnicity, and Immigrant Status

These findings underscore the importance of developing gender-specific and culturally responsive strategies to enhance parent-worker relationships.

Comparative Analysis of Three Facial Scanners for Creating Digital Twins by Focusing on the Difference in Scanning Method

The findings from this study can be utilized in creating digital twins, selecting and merging data, and developing new scanners to overcome limitations.

Voltage-Gated Na+ Channels in Alzheimer's Disease: Physiological Roles and Therapeutic Potential
