Identification of RNA silencing components in soybean and sorghum

Background: RNA silencing is a process triggered by 21-24 small RNAs to repress gene expression. Many organisms including plants use RNA silencing to regulate development and physiology, and to maintain genome stability. Plants possess two classes of small RNAs: microRNAs (miRNAs) and small interfering RNAs (siRNAs). The frameworks of miRNA and siRNA pathways have been established in the model plant, Arabidopsis thaliana (Arabidopsis). Results: Here we report the identification of putative genes that are required for the generation and function of miRNAs and siRNAs in soybean and sorghum, based on knowledge obtained from Arabidopsis. The gene families, including DCL, HEN1, SE, HYL1, HST, RDR, NRPD1, NRPD2/NRPE2, NRPE1, and AGO, were analyzed for gene structures, phylogenetic relationships, and protein motifs. The gene expression was validated using RNA-seq, expressed sequence tags (EST), and reverse transcription PCR (RT-PCR). Conclusions: The identification of these components could provide not only insight into RNA silencing mechanism in soybean and sorghum but also basis for further investigation. Summary: Potential proteins from three novel food sources were predicted and evaluated for allergic cross-reactivity risks by comparing amino acid sequences against allergens in the AOL database. Despite many proteins exceeding CODEX limits for identity matches, these matches were not predictive of allergy risks based on observed taxonomic cross-reactivity. New methods or changes in allergen databases are recommended for better risk evaluation.

Genome-wide discovery of natural variation in pre-mRNA splicing and prioritising causal alternative splicing to salt stress response in rice

Chemical and genetic variation in feral Cannabis sativa populations across the Nebraska climate gradient

Summary: This study investigates the phytochemistry and genomics of feral Cannabis populations collected from Nebraska. It found that both male and female flower tissues produced cannabinoids, with cannabidiol being the primary cannabinoid. The expression of genes related to cannabinoid biosynthesis was higher in female flowers, suggesting transcriptional control of cannabinoid abundance. DNA sequencing revealed that the feral Cannabis plants from Nebraska are more similar to hemp-type Cannabis plants than drug-type plants. This research provides foundational knowledge for future studies of feral Nebraska Cannabis.

Summary: Some crop wild relatives have the ability to tolerate extreme stress beyond what domesticated crops can withstand. This study focuses on Paspalum vaginatum, a self-incompatible and stress-tolerant wild relative of maize and sorghum, and explores the molecular mechanisms that enable it to adapt to nutrient-deficit conditions. The findings suggest that inducing trehalose accumulation in maize, similar to the metabolic phenotype of Paspalum vaginatum, can lead to increased biomass accumulation via autophagy.

The association of blood ctDNA levels to mutations of marker genes in colorectal cancer
