Summary: Monocytes are effective at improving engraftment in HSPC transplantation, particularly in cases of limited cell doses such as CB transplantation and HSC gene therapy. Co-culturing monocytes increases survival and stemness of CB-CD34(+) cells. Soluble factors and direct-cell contact interactions play critical roles in the HSC-monocyte crosstalk.

Summary: This study reveals the binary regulation of y817 cells by microbiota, where microbiota instruct in situ IL-17 production and concomitant expression of the inhibitory receptor PD-1. Microbiota-driven PD-1 inhibits in situ IL-17 production by mucosa-resident y817 effectors, linking microbiota to their simultaneous activation and suppression.

Summary: This study aims to explore the persistent synergistic effects of allogeneic umbilical cord blood (UCB) cells and erythropoietin (EPO) in treating children with cerebral palsy (CP). The results indicate that, compared to baseline scores, all groups showed improvements in outcome measures after 12 months of additional UCB infusion, except for GMFM and GMPM in Group C, which were elevated at 3 and 6 months post-therapy. However, there were no significant differences in outcome measures between the four groups at 3, 6, and 12 months after therapy. Severe dysfunction patients in Group A showed greater improvement in GMPM scores compared to Group D (Ps < 0.05) from the baseline value of the previous trial.

Summary: This study investigated the effect of CD3 signaling on cord blood gamma delta T-cell proliferation. The results showed that stimulation with anti-CD3 antibody significantly increased the population of V delta 1+ T cells, which exhibited the highest cytotoxicity against U937 cells. Additionally, V delta 1+ T cells responded to U937 cells by secreting cytotoxic cytokines such as Granzyme B, IFN-gamma, and sFasL, which correlated with the expression level of PVR and Nectin on target cells.

Summary: This study found that hematopoietic stem/progenitor cells derived from mobilized peripheral blood and bone marrow have similar gene repair capabilities in ex vivo gene therapy for pediatric Wiskott-Aldrich syndrome. However, the peripheral blood group showed better treatment outcomes, with faster recovery and higher numbers of engrafted clones and gene corrections in the myeloid lineage, which are related to the higher amounts of primitive and myeloid progenitors in the peripheral blood-derived cells. This finding provides important references for the clinical interpretation of hematopoietic stem/progenitor cell transplantation outcomes.

Summary: High levels of ICAM-1 in tumors and plasma are associated with prolonged survival in non-small cell lung cancer patients treated with anti-PD-1 or anti-PD-L1. Further studies reveal that sICAM-1 enhances the efficacy of anti-PD-1 by activating cytotoxic T cells, and CXCL13 might be involved in the ICAM-1-mediated anti-tumor pathway. Combinatorial therapy with sICAM-1 and anti-PD-1 shows promise in converting anti-PD-1-resistant tumors to responsive ones in preclinical studies.

Summary: Gamma delta T cells play a crucial role in controlling viral infections, particularly in immunocompromised patients after stem cell transplantation. Recent findings suggest that these cells exhibit an adaptive-like immune response to cytomegalovirus (CMV), with long-lasting clonal expansion and effector functions. It is proposed that gamma delta TCRs are triggered by CMV-induced self-antigens rather than viral antigens. Moreover, the cross-reactivity of CMV-activated gamma delta TCRs with tumor cells makes them promising tools for antiviral and antitumor therapy.

Summary: Inhibition of HIF-1 alpha transcriptional activity can increase levels of NK and CD8(+) T cells in the tumor microenvironment, enhancing the efficacy of cancer immunotherapy. Combining acriflavine can significantly improve the benefits of cancer immunotherapy based on TRP-2 peptide vaccination and anti-PD-1 blocking antibody, while melanoma patients with high CCL5 levels have a higher survival rate.

Summary: The reduction of human CD1c+CD5+ dendritic cells (DCs) in melanoma-affected lymph nodes and the correlation between CD5 expression on DCs and patient survival have been observed. Activating CD5 on DCs enhances T cell priming and improves survival after immune checkpoint blockade (ICB) therapy. CD5+ DCs are essential for optimal ICB therapy.
