Summary: In this study, the full metagenome sequences from the soil of Tuwa hot-spring in Gujarat, India were examined for the presence of thermostable enzymes. The a-amylase gene was found, amplified, cloned, and expressed in Escherichia coli. The rm-a-amylase showed high activity at 60 ℃, pH 6.0, and in the presence of heavy metal ions, and it can hydrolyze agar, xylan, pectin, alginate, and cellulose.

Summary: A new carrageenase gene, car1383, was found from the metagenome of Antarctic macroalgae-associated bacteria. The recombinant Car1383 showed activity in degrading carrageenan into neocarrabiose and neocarratetraose. The study also identified the optimal activity conditions and important structural sites of Car1383. This research provides a new candidate for the industrial production of bioactive algal oligosaccharides.

Summary: This study isolated and identified a thermophilic cellulolytic bacterium from hot spring water and characterized its thermostable cellulase. The bacterium, identified as Geobacillus sp. KP43, showed optimal growth between 55°C and 65°C and produced cellulase enzyme with a molecular weight of approximately 66 kDa. The enzyme exhibited optimal activity at 70°C and a wide pH range and was stimulated by Fe2+ ions. The bacterium also displayed good activities of xylanase, cellobiase, and amylase. The findings suggest that Geobacillus sp. KP43 can withstand harsh environmental conditions and is suitable for enzyme production in various biotechnological and industrial applications.

Summary: In this study, the iota-carrageenase gene Car1293 was obtained from Microbulbifer sp. YNDZ01, and its sequence, protein structure, enzymatic properties, enzymatic digestion products, and anti-inflammatory activity were investigated. The results showed that Car1293 encodes a novel enzyme that can hydrolyze carrageenan into CGOS-DP8 with significant anti-inflammatory activity. This study fills a gap in the research on the biological activity of oligosaccharides in iota-carrageenan and provides promising data for the development of natural anti-inflammatory agents.

Summary: A novel xylanase gene, xynDRTY1, was identified from Tengchong hot spring using a metagenomic approach. The gene was codon-optimized, synthesized, and overexpressed in Escherichia coli. The purified XynDRTY1 showed activity with various natural glycosides, exhibited optimal pH and temperature, and demonstrated stability, making it potentially useful for biotechnological applications.

Summary: In the last two decades, researchers have focused on studying the diverse microorganism-based communities found in natural hot springs, particularly thermophiles, to explore their potential applications in various industrial and biotechnological fields. This study isolated 83 thermophilic Bacilli strains from different geothermal hot springs in Turkey and characterized their physiological, morphological, biochemical, and molecular properties. The results revealed the presence of five different species and demonstrated the production potential of hydrolytic enzymes among the isolates. The findings contribute to our understanding of the microbial diversity and enzyme-producing capabilities of thermophiles in natural hot spring environments.

Summary: A new thermophilic species of the genus Allochromatium, named Allochromatium tepidum NZ(T), was described in this study. This bacterium was isolated from a microbial mat in the sulfidic geothermal outflow of a hot spring in Rotorua, New Zealand and grew optimally near 45 degrees C. It had a similar carotenoid composition as Thermochromatium tepidum and grew photoheterotrophically on acetate in the absence of sulfide, unlike other thermophilic purple sulfur bacteria.

CeCgkB exhibited cold-adapted properties and high substrate affinity, and it could degrade x-carrageenan into products of varying DP. The major products of CeCgkB were identified, and its potential application in the efficient preparation of x-carrageenan oligosaccharides was demonstrated.

Summary: A novel lipase gene (lip4346) was identified from the genome of a macroalgae-associated bacterial strain, cloned, and heterologously expressed in Escherichia coli. The purified Lip4346 protein showed high enzyme activity, optimal temperature at 70 degrees Celsius, and tolerance towards various solvents and detergents, making it a promising nonaqueous biocatalyst for industrial applications.
