Summary: This study investigated the solubility of fully glycosylated and non-glycosylated human monoclonal antibodies (mAb) expressed in corn by inducing precipitation with poly(ethylene) glycol (PEG). The findings show that glycosylation increases antibody solubility and that existing models cannot accurately capture the effect of glycosylation on protein precipitation by PEG.

Summary: A continuous mAb platform was developed with both complete continuous (CC) and periodic continuous (PC) modes of operation, showing that CC mode had higher specific productivity and lower consumable utilization, while PC mode was operationally simpler and had lower facility costs due to reduced auxiliary equipment. Both modes offered viable options for continuous processing of mAbs and achieved target critical quality attribute profiles of the final drug product within 24 hours of operation.

The method involved extraction of trastuzumab from serum samples, followed by reduction, alkylation, trypsin digestion, and quantification using LC-MS/MS. The assay provided a novel approach for accurate and quantitative monitoring of mAb-trastuzumab.

Summary: In this study, a platform for purifying therapeutic antibodies via affinity precipitation using the endogenous calcium ion-binding protein, calsequestrin (CSQ), is reported. The ZZ-CSQ fusion proteins capture antibodies and undergo multimerization and subsequent aggregation in response to calcium ions, enabling the antibody to be collected by affinity precipitation. The platform is rapid, recyclable, scalable and cost-effective, and has better impurity removal performance compared to traditional protein A-based affinity chromatography.

Summary: This study examines the purification of Trastuzumab using cation-exchange z(2) laterally-fed membrane chromatography (z(2)LFMC). The results show that the z(2)LFMC process is comparable to protein A affinity chromatography in terms of purity and recovery, but it is faster and does not require acidic conditions. The productivity of Trastuzumab with the z(2)LFMC process is more than 3 times higher than with column-based purification. Therefore, z(2)LFMC could be a cheaper and more efficient alternative to protein A affinity chromatography.

Summary: The use of precipitation as a low-cost alternative to Protein A chromatography has gained renewed interest due to increased product titers and the focus on continuous bioprocessing. In this study, a commercially relevant monoclonal antibody (mAb) was purified from cell culture fluid using a tubular flow precipitation reactor and tangential flow microfiltration for dewatering and washing. The results showed high mAb yield, DNA removal, and comparable host cell protein clearance, making precipitation a promising primary capture method for mAb products.

Summary: This study investigates the improved clarification of cell culture fluids for the production of monoclonal antibodies through precipitation and flocculation pretreatments, combined with a mild fluidized bed centrifuge separation and filtration step. The findings show that these pretreatments enhance the filtering capacity and achieve high mAb recoveries, low turbidities, and high DNA removals, suggesting consistent mAb quality.

Summary: In this study, a coupled process combining ion exchange chromatography and polyethylene glycol precipitation was developed to remove acidic charge variants of monoclonal antibodies. The model evaluation showed that the coupled process outperformed stand-alone ion exchange chromatography, with the improvement in throughput depending on the initial content and reduction demand of the acidic variants.
