Novel molecular hybrids of cinnamic acids and guanylhydrazones as potential antitubercular agents

In an attempt to identify potential new agents active against tuberculosis, 20 novel phenylacrylamide derivatives incorporating cinnamic acids and guanylhydrazones were synthesized using microwave assisted synthesis. Activity of the synthesized compounds was evaluated using resazurin microtitre plate assay (REMA) against Mycobacterium tuberculosis H37Rv. Based on empirical structure-activity relationship data it was observed that both steric and electronic parameters play major role in the activity of this series of compounds. Compound 7s (2E)-N-((-2-(3,4-dimethoxybenzylidene) hydrazinyl) (imino) methyl)-3-(4-methoxyphenyl) acrylamide showed MIC of 6.49 mu M along with good safety profile of >50-fold in VERO cell line. Thus, this compound could act as a potential lead for further antitubercular studies. (C) 2010 Elsevier Ltd. All rights reserved.

Production, characterization and in-vitro applications of single-domain antibody against thyroglobulin selected from novel T7 phage display library

Summary: Single-domain antibodies (SdAbs) have been used in various biomedical applications, but their use in immunoradiometric assays (IRMA) for thyroglobulin (Tg) has not been reported. This study demonstrates the production of SdAbs against human-Tg using a hybrid immunization/directed-evolution approach. The T7 phage display system shows potential as an alternative for constructing single-domain antibody display libraries. Additionally, the SdAbs showed quantitative-binding properties in IRMA for thyroglobulin, with serum Tg values correlating significantly with those obtained using conventional methods.

Summary: This study isolated, identified, and evaluated the safety of two novel L. reuteri strains. Genomic sequencing and in vitro assays confirmed their species identity, absence of known genetic sequences of concern, susceptibility to antibiotics, and lack of harmful by-products. In vivo experiments using a bird model demonstrated the safety of these strains. These findings provide evidence for their potential probiotic applications in humans and animals.

Summary: Antibiotic resistance is a major threat to public health, with Gram-negative bacteria presenting unique challenges due to their low permeability and efflux pumps. Limited understanding of the chemical rules for overcoming these barriers hinders antibacterial drug discovery. Efforts to address this issue, such as screening compound libraries and using cheminformatic analysis, have led to the design of sulfamidoadenosines with diverse substituents, showing potential utility in accumulation in Escherichia coli.

Design of MERS-CoV entry inhibitory short peptides based on helix-stabilizing strategies

Summary: This study successfully developed a short peptide mimetic as a MERS-CoV fusion inhibitor by reproducing the key recognition features of the HR2 helix. The resulting 23-mer lipopeptide showed comparable inhibitory effect to the 36-mer HR2 peptide HR2P-M2. This has important implications for developing short peptide-based antiviral agents to treat MERS-CoV infection.

Summary: Beta(2)-adrenergic receptor agonists can stimulate glucose uptake by skeletal muscle cells and are therefore potential treatments for type 2 diabetes. The chirality of compounds has a significant impact on the activity of these agonists. This study found that certain synthesized compounds showed higher glucose uptake activity. These findings provide important information for the design of novel beta(2)AR agonists for T2D treatment.

Conformationally constrained potent inhibitors for enhancer of zeste homolog 2 (EZH2)

Summary: The study focuses on EZH2, a promising therapeutic target for various types of cancers. Researchers designed and synthesized a series of novel derivatives aiming to enhance the EZH2 inhibition activity. Among them, compound 28 displayed potent EZH2 inhibition activity and showed high anti-proliferative effects in lymphoma cell lines and xenograft mouse models.
