Summary: Cu@PB NCs exhibit enhanced POD-like activity compared to PB NCs and HRP, and can be utilized for colorimetric assay of ALP activity by oxidizing TMB in the presence of H2O2. Introduction of PPi results in decreased POD-like activity of Cu@PB NCs, while ALP hydrolyzes PPi and restores the activity of Cu@PB NCs.

Summary: Little is known about the biological roles of the protein histidine phosphatase PHPT1, and the lack of chemical tools for studying its phosphorylation and dephosphorylation has hindered progress in the field. Through an inhibitor screen using a fluorescent assay, norstictic acid was identified as a potent inhibitor of PHPT1 activity.

Summary: A sensing platform was developed for determining alkaline phosphatase activity based on the cooperation of DNAzyme-Au spherical nucleic acid nanoprobe with the graphene-oxide-loaded hybridization chain reaction system. The assay utilizes the strong affinity between pyrophosphate and Cu2+ ions, as well as the hydrolysis of pyrophosphate by ALP to release Cu2+ ions, to achieve good detection sensitivity and specificity. The feasibility of the method was validated by spiking ALP standards in bovine serum, showing potential for drug screening applications.

Summary: This work presents the development and characterization of fluorescent nanocomposite hydrogels using a green protocol. The hydrogels have high swelling and absorption capacity and are prepared by incorporating polyfluorenes-based nanoparticles into a three-dimensional polymeric network. The hydrogels show good storage stability and homogeneity when using the ex situ method. They have been tested as a sensing platform for the detection of alkaline phosphatase with a quick response time and low LOD, making them promising for quantifying physiological ALP levels and enzyme activity in environmental samples.

Summary: In this study, a trienzyme cascade-triggered fluorescent immunosensor platform was constructed by integrating alkaline phosphatase (ALP), tyrosinase (TYR), and horseradish peroxidase (HRP). The platform utilized a rapid in situ fluorogenic reaction to produce a yellow fluorescent compound. The developed immunosensor showed significant enhancements in fluorescence and successfully detected a cardiac biomarker in clinical samples.

Summary: Phosphatase is crucial in many biological functions, with alkaline phosphatase (ALP) serving as a key prognostic indicator for various diseases, though current detection methods are limited to in vitro colorimetric techniques. Fluorescence imaging provides a reliable way to monitor real-time changes in ALP levels, with research focusing mainly on ALP imaging probes grouped by their luminescence mechanisms. Challenges and future prospects for phosphatase probes were also discussed.

Summary: The purpose of this meta-analysis was to summarize the diagnostic value of serum alkaline phosphatase (ALP) and bone-specific isoforms (BAP) in metastatic breast cancer (BC), with a focus on bone metastases. High levels of ALP and BAP in serum were found to be associated with a higher detection rate of bone metastatic BC patients. Although not highly reliable, serum ALP and BAP still provide some useful information for early detection of BC metastases, especially bone metastases.

Summary: A facile hydrothermal method was developed to prepare FeWO4 nanomaterials with intrinsic peroxidase-like activity, and the catalytic activity was significantly enhanced by adjusting the size and oxygen vacancy content. A colorimetric assay based on FeWO4 was constructed to detect PPi and ALP activity. The proposed method successfully evaluated ALP activity in real serum samples and screened for potential ALP inhibitors.
