Summary: The study confirmed that miR-137 directly targets the 3 ' UTR of NOTCH1 while positively regulates HES1 expression. Additionally, it was found that tangeretin can reverse the effects of miR-137 knockdown on osteogenic promotion and downstream gene expression. Furthermore, a co-regulatory signaling network of NOTCH1/LSD1/BMP2 was proposed to elucidate the modulation of miR-137 on the osteoblastic differentiation of hASCs.

Summary: Exosomes derived from human adipose-derived stem cells (hADSCs) can preferentially accumulate at bone, especially at the calvarial defect site, when intravenously injected. Exosomes treatment promotes osteogenesis and bone defect repair. Moreover, exosomes from 3D-cultured hADSCs exhibit superior bone-targeting effects and repair capacity compared to those derived from 2D-cultured hADSCs.

Summary: This paper presents the design of a tough and flexible amphoteric copolymer-based hydrogel for bone regeneration, which shows good osteogenesis induction and osseointegration. The hydrogel is copolymerized with N-acyl glycinamide (NAGA), anionic acrylate alendronate (AcAln), and cationic (2-(acryloyloxy)ethyl) trimethyl ammonium chloride (DMAEA-Q), and contains three types of physical cross-links. In vitro and in vivo experiments demonstrate the excellent osteogenic properties of the hydrogel.

Summary: In this study, PRP was encapsulated in a cryogel to address issues such as burst release of growth factors and activation with exogenous activators. The prepared PRP-cryogel effectively saved autologous activators while reducing the release rate of growth factors, enhancing the long-term osteogenesis of ADSCs. The combination of PRP, cryogel, and ADSCs provides temperature-activated PRP and slow-release growth factors for bone repair in rats.

Summary: Adipose-derived stem cells (ADSCs) in 3D-cultured spheroid form promote bone regeneration, suppress cell apoptosis, and elevate gene expression related to bone morphogenesis, angiogenesis, and stemness. Injectable small size ADSCs spheroids are a novel and less-invasive therapeutic option for treating bone defects.

Summary: Prx1 expression is not only found in bone marrow and periosteum, but also in muscle, promoting ectopic bone formation. This study investigated the regulatory mechanisms and differentiation potential of Prx1-SSCs derived from muscle and periosteum. It was found that muscle-derived Prx1-SSCs are quiescent and less responsive to BMP2 compared to periosteal Prx1-SSCs.

Summary: This study investigated the effects of oxytocin on the proliferation, migratory ability, senescence, and autophagy of human adipose-derived stem cells (hASCs). The results showed that oxytocin did not affect hASC proliferation and migratory ability, but promoted autophagy and osteogenic differentiation. This suggests that oxytocin may promote osteogenesis in hASCs by modulating the autophagic process.

Summary: The study investigated the enhancement of osteogenic and angiogenic potential of Exosomes through overexpression of exosomal miRNA (microRNA) and the long-term effects of Exos loaded in HMPs (Hydrogel microparticles) on new bone formation. The research demonstrated that miR-29a overexpression inhibited HDAC4 and upregulated RUNX2 and VEGF, enhancing the osteogenic and angiogenic capabilities. The synthesized HMP@Exo system showed good injectability, compatibility, and cell adhesion properties, with sustained release of Agomir-29a-Exos enhancing cell proliferation, migration, osteogenesis, and angiogenesis. Overall, the injectable HMP@Exo system provides an innovative approach for coupling osteogenesis and angiogenesis, promoting fracture healing.

Summary: This study aimed to compare the intraoperative hemodynamic changes between the wide-awake local anesthesia no tourniquet (WALANT) technique and general anesthesia (GA) in patients undergoing distal radius plating surgery. The results showed that the WALANT group had significantly decreased pain intensity, while there were no significant changes in intraoperative mean arterial pressure (MAP). Additionally, the WALANT group had fewer MAP fluctuations compared to the GA group. The reduction and plating quality were similar between the two groups.

Summary: We investigated the apoptotic response of glioblastoma cells to different drugs and found that p53 is associated with DNA damage and apoptosis initiation. The transcriptional activity of p53 does not play a role in doxorubicin-induced apoptosis, but p53 concentrates in the nucleus during heavy apoptosis. Bortezomib is sensitive to cell apoptosis, and vorinostat reduces p53 levels in U87 cells. The combination of doxorubicin and bortezomib induces compound stress, resulting in synergistic apoptosis in U87 cells, without involvement of p53.

Summary: This paper reviews the recent developments of two newly developed intermetallic compounds as potential implantable biomaterials. The high strength, low Young's modulus, and excellent corrosion resistance make these compounds good candidates for bio-implantable materials. Additive manufacturing techniques provide an advantageous route for the preparation of these materials. A practical example confirms the feasibility of additive manufacturing for producing implantable intermetallic compound-based prostheses.

Summary: This study aims to establish a surface modification technology for ZK60 magnesium alloy implants that can degrade uniformly over time and promote bone healing. It proposes a special micro-arc oxidation (MAO) treatment on ZK60 alloy to create a coating with better corrosion resistance and solve the problems of uneven and excessive degradation. The MAO-treated samples exhibited excellent corrosion resistance and electrochemical stability, and the deposited Ca ions were not cytotoxic and were beneficial for enhancing bone healing after implantation.
