Summary: This study uncovers the transcriptional network controlled by lipolysis in brown adipocytes and shows that lipolysis is required for the activation of the thermogenic gene program by beta-adrenergic signals. PPARs are identified as key mediators of lipolysis-induced activation of lipid metabolism and thermogenic genes. Additionally, lipolysis also activates the unfolded protein response and regulates the core circadian transcriptional machinery independently of PPARs.

Summary: This study presents the cryo-EM structures of human GPAT1 in substrate analog-bound and product-bound states, revealing the important catalytic motifs and critical C-terminal domain for proper protein folding. Unexpectedly, GPAT1 associates with the membrane via an amphipathic surface patch and an N-terminal loop-helix region, and has a hydrophobic pathway for lipid trafficking. These findings lay a framework for rational inhibitor development for GPAT1.

Summary: Brown adipocytes are specialized fat cells that dissipate energy as heat instead of ATP synthesis. These cells have the unique ability to oxidize substrates independent of ADP availability and preferentially oxidize free fatty acids released from lipid droplets to produce heat during cold exposure. Additionally, brown adipocytes can take up large amounts of glucose and increase glycolysis and fatty acid synthesis. This review summarizes mechanisms regulating mitochondrial substrate selection and discusses recent findings on two distinct populations of brown adipocyte mitochondria with different substrate preferences. The review further explores how these mechanisms allow for simultaneous increases in glycolysis, fatty acid synthesis, and fatty acid oxidation in brown adipocytes.

Summary: We found that the decreased expression of YB-1 in the BAT of aged mice is due to the reduction of the microbial metabolite butyrate. Overexpression of YB-1 in the BAT of aged mice can promote BAT thermogenesis and alleviate diet-induced obesity and insulin resistance. Additionally, we discovered that the natural compound Sciadopitysin can enhance YB-1 protein stability and nuclear translocation, improving BAT aging and metabolic disorders.

Summary: Species of Schizochytrium are well known for their ability to produce lipids intracellularly. In this study, seven different natural antioxidants were evaluated for their effects on lipid accumulation in Schizochytrium sp. PKU#Mn4. Mannitol showed the best increment in total fatty acids concentration, with interactions between mannitol and ascorbic acid resulting in increased saturated and polyunsaturated fatty acids concentrations. This study provides insights into strategies for improving lipid accumulation in thraustochytrids using exogenous antioxidants and the underlying mechanisms.

Summary: This study identified BnaGPAT9 as the main ortholog of GPAT9 in Brassica napus, and found that its overexpression promotes seed oil accumulation and eukaryotic galactolipid synthesis. BnaGPAT9 is highly expressed in developing seeds and localized in the endoplasmic reticulum. By increasing phosphatidic acid production and altering lipid composition, BnaGPAT9 enhances seed oil accumulation.

Summary: The study identified a class of hypotriglyceridemic omega-3 fatty acid-derived N-acyl taurines (NATs) in bile that can inhibit intestinal triacylglycerol hydrolysis and lipid absorption, preventing hypertriglyceridemia and fatty liver. These omega-3 NATs may contribute to the mechanism of action of fish oil and influence the design of more potent omega-3 fatty acid-based therapeutics.

Summary: This study investigated the physiological function of BAAT in bile acid and lipid metabolism using Baat(-/-) mice. The results showed that BAAT deletion affected bile acid synthesis and metabolism, altered energy metabolism and microbiome structure in mice. Furthermore, when fed a westernized diet, Baat(-/-) mice showed compromised weight gain and impaired insulin secretion.
