Autophagy-associated release of HMGB1 is known to protect cancer cells from many chemotherapeutics. However, the detailed molecular mechanism(s) responsible remains largely undefined. We show in this study that HMGB1 released into the extracellular space protects gastric cancer cells from apoptosis induced by the microtubule-targeting drug vincristine through transcriptional upregulation of Mcl-1. Extracellular HMGB1 appeared essential for autophagy-mediated inhibition of apoptosis, in that siRNA knockdown of HMGB1 or inhibition of its release abolished the protective effect of autophagy. Strikingly, vincristine upregulated the Mcl-1 mRNA expression through a transcriptional increase, but did not alter the expression levels of the Mcl-1 protein. Inhibition of HMGB1 release blocked the increase in the Mcl-1 transcript and caused reduction in Mcl-1 at the protein level, indicating that HMGB1-mediated signaling was necessary for transcriptional upregulation of Mcl-1. This seemed critical for maintaining sufficient Mcl-1 protein expression required for survival of gastric cancer cells exposed to vincristine. The effect of extracellular HMGB1 on transcriptional regulation of Mcl-1 was confirmed in gastric cancer cells treated with recombinant HMGB1. Taken together, these results identify HMGB1-mediated upregulation of Mcl-1 transcription as an important mechanism by which autophagy protects gastric cancer cells from apoptosis induced by vincristine.

Repurposing Melanoma Chemotherapy to Activate Inflammasomes in the Treatment of BRAF/MAPK Inhibitor Resistant Melanoma

Farzana Ahmed, Hsin-Yi Tseng, Antonio Ahn, Dilini Gunatilake, Sara Alavi, Michael Eccles, Helen Rizos, Stuart J. Gallagher, Jessamy C. Tiffen, Peter Hersey, Abdullah Al Emran

Summary: This study explores the resistance of melanoma to chemotherapy and suggests that melanoma with an inflammatory undifferentiated state may be susceptible to combination therapy. The study also investigates the variation in inflammasomes and death mechanisms in different melanoma, and raises the possibility of using the inflammatory state as biomarkers for inflammasome-targeted therapy.

The long noncoding RNA glycoLINC assembles a lower glycolytic metabolon to promote glycolysis

Youming Zhu, Leu Jin, Ronghua Shi, Jinming Li, Yan Wang, Li Zhang, Chao-Zhao Liang, Vinod K. Narayana, David P. De Souza, Rick F. Thorne, Li Rong Zhang, Xu Dong Zhang, Mian Wu

Summary: This study reveals a novel long noncoding RNA (lncRNA) called gLINC that acts as a backbone for the formation of a metabolon involving glycolytic enzymes and lactate dehydrogenase A. The gLINC metabolon enhances glycolytic flux, ATP production, and cell survival under serine deprivation. Overexpression of gLINC in cancer cells promotes xenograft growth in mice on a serine-deprived diet, suggesting its role in cancer cell adaptation.

Summary: This study explores the role of nicotinamide in preventing and reversing T cell exhaustion, which has important implications for immunotherapy against cancers and viral infections. Nicotinamide supplementation was found to prevent the development of inhibitory receptors in T cells and promote the differentiation of T cells to effector states, suggesting a potential therapeutic strategy for enhancing immune responses.

Summary: There is increasing evidence suggesting the involvement of nerve growth factor (NGF) and its receptors in cancer growth. In this study, the expression of NGF, proNGF, TrkA, p75(NTR) and sortilin in melanoma was investigated. The results showed downregulation of NGF/ProNGF and TrkA in melanoma, indicating a potential therapeutic target.

Glycolytic Pfkp acts as a Lin41 protein kinase to promote endodermal differentiation of embryonic stem cells

Leixi Cao, Ruijie Wang, Guangzhi Liu, Yuwei Zhang, Rick Francis Thorne, Xu Dong Zhang, Jinming Li, Yang Xia, Lili Guo, Fengmin Shao, Hao Gu, Mian Wu

Summary: This study reveals an interaction between LIF-Stat3 signaling and the glycolytic enzyme Pfkp in the differentiation of murine ESCs into specific cell lineages. The repression of Pfkp by Stat3 maintains pluripotency in ESCs, and lifting this repression results in differentiation towards the ectodermal lineage. Pfkp acts as a protein kinase to phosphorylate Lin41, stabilizing its expression and promoting the binding and degradation of mRNAs encoding ectodermal specification markers.

Combining Neuropsychological Assessment with Neuroimaging to Distinguish Early-Stage Alzheimer's Disease from Frontotemporal Lobar Degeneration in Non-Western Tonal Native Language-Speaking Individuals Living in Taiwan: A Case Series

Chih-Yun Kuo, Hsin-Yi Tseng, Ivo Stachiv, Chon-Haw Tsai, Yi-Chun Lai, Tomas Nikolai

Summary: Neuropsychological tests are widely used for dementia assessment in clinical practice and are also important for differentiating between Alzheimer's disease (AD) and frontotemporal lobar degeneration (FTLD), especially in the initial clinical presentations of behavioral variants of frontotemporal dementia (bvFTD) and primary progressive aphasia (PPA). However, the overlapping signs and cultural differences raise concerns about the validity and reliability of these tests. This case series combined NPTs with neuroimaging to examine their effectiveness in distinguishing between AD and FTLD, and the results showed that participants diagnosed with FTLD had lower scores in language and social cognition tests compared to AD participants, while PPA participants had lower scores in the Free and Cued Selective Reminding Test compared to bvFTD participants.

Grey-Markov model of user demands prediction based on online reviews

Na Zhang, Ling Qin, Ping Yu, Wei Gao, Yupeng Li

Summary: With the development of e-commerce and the increase in online shopping, users have higher demands on products. Enterprises need to design products that meet user demands accurately and quickly in order to improve user satisfaction and product competitiveness. This paper proposes a Grey-Markov model of user demands prediction based on online reviews, which predicts the monthly changing trend of user demands in advance. By using LDA topic model and sentiment analysis, user attention values and satisfaction values are obtained. The effectiveness of the proposed method is illustrated through examples of demand prediction and optimization design for smartphones and automobiles, providing a reference for manufacturing enterprises to optimize product design.
