Objective-Suppressors of cytokine signaling (SOCS) proteins are intracellular regulators of receptor signal transduction, mainly Janus kinase/signal transducers and activators of transcription (JAK/STAT). We investigated the effects of SOCS modulation on the JAK/STAT-dependent responses in vascular cells, and their implication in atherosclerotic plaque development. Methods and Results-Immunohistochemistry in human plaques revealed a high expression of SOCS1 and SOCS3 by vascular smooth muscle cells (VSMCs) and macrophages in the inflammatory region of the shoulders, when compared to the fibrous area. SOCS were also increased in aortic lesions from apoE(-/-) mice. In cultured VSMCs, endothelial cells, and monocytes, SOCS1 and SOCS3 were transiently induced by proinflammatory cytokines, proatherogenic lipoproteins, and immune molecules. Furthermore, overexpression of SOCS suppressed STAT activation and reduced inflammatory gene expression and cell growth, whereas SOCS knockdown increased these cell responses. In vivo, antisense oligodeoxynucleotides targeting SOCS3 exacerbated the atherosclerotic process in apoE(-/-) mice by increasing the size, leukocyte content, and chemokine expression in the lesions. Conclusions-SOCS expressed in atherosclerotic lesions are key regulators of vascular cell responses. Activation of this endogenous antiinflammatory pathway might be of interest in the treatment of atherosclerosis. (Arterioscler Thromb Vasc Biol. 2009; 29: 525-531.)

Beneficial effects of an algal oil rich in ω-3 polyunsaturated fatty acids on locomotor function and D2 dopamine receptor in haloperidol-induced parkinsonism

Alejandra Barroso-Hernandez, Abril Ramirez-Higuera, Carolina Pena-Montes, Sergio Alberto Cortes-Ramirez, Mauricio Rodriguez-Dorantes, Oscar Lopez-Franco, Rosa Maria Oliart-Ros

Summary: This study investigated the effects of an omega-3 PUFA rich algal oil on locomotive alterations induced by haloperidol and D-2 receptor protein and gene expression in Wistar rats. The results confirmed the beneficial effects of omega-3 PUFA in reducing locomotive alterations and stimulating D-2 receptor presence, indicating their positive role in brain health.

Summary: This study compared the prognostic value of 3 different LN classifications for patients with resected GC, showing that LNR-based classification stratified patients better for prognosis assessment. Further studies are needed to evaluate the impact of the number of LN examined, cutoff values, and anatomic extent of LN disease in GC.

Summary: Analyzing the tumor-LNR classification can successfully predict the prognosis of resected gastric cancer patients. The T-LNR classification is significantly associated with tumor characteristics, patient progression, and death, demonstrating good stratification and discrimination capabilities.

Summary: This study identified a six-gene predictive signature of trabectedin efficacy in advanced soft-tissue sarcomas. By evaluating 118 genes involved in DNA damage repair, potential biomarkers of trabectedin sensitivity or resistance were defined. Targeting some genes of this signature may enhance trabectedin efficacy in treatment.

Cristina Diaz del Arco, Luis Ortega Medina, Lourdes Estrada Munoz, Elena Molina Roldan, M. Angeles Ceron Nieto, Soledad Garcia Gomez de las Heras, M. Jesus Fernandez Acenero

Summary: Borrmann classification is commonly used for advanced gastric cancer diagnosis. The classification shows distinct clinicopathological and biological entities in patients, but does not have a direct association with prognosis. Further research is needed to verify its role in patient stratification.

Summary: Intestinal metaplasia is associated with an increased risk of gastric cancer, and finding molecular biomarkers can aid in guiding prevention programs. Helicobacter pylori infection influences DNA methylation, requiring early adoption of prevention strategies. Research suggests that intestinal metaplasia partially resembles the methylation patterns of intestinal gastric cancer.
