Significance: Proteins destined to secretion and exposure on the cell surface are synthesized and processed in the extracellular-like environment of the endoplasmic reticulum (ER) of higher eukaryotic cells. Compartmentation plays a crucial role in the post-translational modifications, such as oxidative folding and N-glycosylation in the ER lumen. Transport of the required intermediates across the ER membrane and maintenance of the luminal redox conditions and Ca2+ ion concentration are indispensable for appropriate protein maturation. Recent Advances: Cooperation of enzymes and transporters to maintain a thiol-oxidizing milieu in the ER lumen has been recently elucidated. Ca2+-dependence of certain ER chaperones is a subject of intensive research. Critical Issues: Mounting evidence supports the existence of a real barrier between the ER lumen and the cytosol. The unique set of enzymes, selection of metabolites, and characteristic ion and redox milieu of the luminal compartment strongly argue against the general permeability of the ER membrane. Future Directions: Alterations in the luminal environment can trigger the unfolded protein response, a common event in a variety of pathological conditions. Therefore, redox and calcium homeostasis and protein glycosylation in the ER provide novel drug-targets for medical treatment in a wide array of diseases. Antioxid. Redox Signal. 16, 1100-1108.

Summary: The study presents a simple and reliable method for the quantification of a group of relevant fatty acids using gas chromatography and flame ionization detection. The method showed good linear calibration and high recovery rates for different types of samples, making it suitable for studying the metabolism of dietary trans fatty acids (TFAs).

Molecular Mechanisms Underlying the Elevated Expression of a Potentially Type 2 Diabetes Mellitus Associated SCD1 Variant

Summary: Disturbances in lipid metabolism caused by excessive food intake and a sedentary lifestyle are major risks for metabolic disorders. The enzyme SCD1 plays an important role in these diseases by catalyzing the synthesis of unsaturated fatty acids. Studies have shown that increased activity or over-expression of SCD1 is a contributing factor for type 2 diabetes mellitus. This research investigated the impact of a common genetic variant on SCD1 function and found that the variant could contribute to the development of obesity-related metabolic disorders, including type 2 diabetes mellitus, by increasing intracellular levels of SCD1.

Synthesis and Antiproliferative Activity of Novel Imipridone-Ferrocene Hybrids with Triazole and Alkyne Linkers

Summary: Imipridones, including ONC201, ONC206 and ONC212, are a novel class of anticancer agents that have shown promising results in clinical trials. In order to enhance the ROS responsivity of these compounds, researchers have designed and synthesized a series of ferrocene-imipridone hybrids. These hybrids exhibited significantly lower cell viability in malignant cell lines compared to ONC201, and two of them were able to overcome resistance and induce apoptosis/necrosis in cells.

Different Metabolism and Toxicity of TRANS Fatty Acids, Elaidate and Vaccenate Compared to Cis-Oleate in HepG2 Cells

Summary: Trans fatty acids (TFAs), ingested in substantial amounts, are believed to be unhealthy and contribute to obesity, cardiovascular diseases, and diabetes. This study used a hepatoma model to compare the metabolism and effects of industrial and ruminant TFAs with that of cis-oleate. The results show that TFAs are less protective against palmitate toxicity compared to oleate, which may be due to their diverse incorporation into accumulating diacylglycerols and ceramides. These findings provide evidence against dietary trans fat intake and help us better understand the molecular mechanisms of lipotoxicity.

Summary: The combined prevalence of type 1 and type 2 diabetes is increasing worldwide. This study investigated the role of the SCD5 promoter and its polymorphisms in diabetes development. The rs3811792 SNP in the SCD5 promoter region was found to reduce promoter activity and was associated with both type 1 and type 2 diabetes.

MEK Is a Potential Indirect Target in Subtypes of Head and Neck Cancers

Summary: The poor prognosis of head-and-neck squamous cell carcinoma (HNSCC) is due to the lack of prognostic and predictive markers. The Ras/Raf/MEK/ERK signaling pathway is activated by overexpressed epidermal growth factor receptor (EGFR) and promotes the progression of HNSCCs. Our study investigated the effects of EGFR and MEK inhibitors on HNSCC-cell lines and found variations in sensitivity among different cell lines. The feedback activation of Akt through EGFR played a role in FaDu cells, while the insensitivity of Detroit 562 cells might be due to a PIK3CA mutation. The versatility of EGFR-initiated signaling pathways in HNSCC cell lines justifies the combined application of EGFR and MEK inhibitors.

Novel Erlotinib-Chalcone Hybrids Diminish Resistance in Head and Neck Cancer by Inducing Multiple Cell Death Mechanisms

Summary: We synthesized novel erlotinib-chalcone molecular hybrids and evaluated their anticancer activity on HNSCC cell lines. The hybrids showed higher efficacy compared to the 1:1 combination of erlotinib and a reference chalcone. They eradicated HNSCC cells in low micromolar concentrations and their mechanism of action was independent of canonical targets. The therapeutic potential of these hybrid molecules validates their development and further investigation is justified to reveal the underlying mechanism.

Molecular Basis of Unequal Alternative Splicing of Human SCD5 and Its Alteration by Natural Genetic Variations

Summary: Alternative splicing is an important post-transcriptional control mechanism that provides protein isoform diversity. Changes in the expression and splicing of SCD5 have significant value for cancer diagnosis, prognosis, and treatment. The study explores the distribution of SCD5 transcript variants in human tissues, the functionality of relevant splice sites, and the influence of single-nucleotide variations. An imbalance in SCD5 isoforms is attributed to weaker recognition of the splicing acceptor site, and natural variations at splice sites can modify the dominance of certain isoforms. These findings provide valuable data on SCD5 in human tissues and reveal mutation-driven changes in alternative splicing, which may impact tumor-associated lipid metabolism and have prognostic implications for novel therapeutic approaches.
