Summary: This study investigated the role and mechanism of Myricitrin (Myr) in regulating synovitis and joint destruction mediated by fibroblast-like synoviocytes (FLS) in rheumatoid arthritis (RA). The results showed that Myr treatment inhibited the migration and invasion of RA FLS, reduced the expression of inflammatory cytokines and MMPs, and suppressed the phosphorylation of AKT. Furthermore, Myr administration relieved arthritis symptoms in a mouse model of collagen-induced arthritis (CIA).

This discovery suggests that NC may be a novel therapeutic agent for RA, controlling FLS-mediated rheumatoid synovial inflammation and joint destruction.

Summary: This study analyzed the specific mechanism of NLRP3 in rheumatoid arthritis (RA). It was found that NLRP3 levels were increased in synovial tissues and fibroblast-like synoviocytes (FLS) of RA patients. Silencing NLRP3 suppressed FLS proliferation and inflammatory factor levels. Additionally, it was discovered that ALKBH5 binds with NLRP3 and its silencing also suppressed FLS proliferation and inflammatory factor levels, while NLRP3 overexpression neutralized the role of ALKBH5 in FLS. Furthermore, m6A modification induced by ALKBH5 suppressed NLRP3 mRNA level through YTHDC2 in RA, and NLRP3 was identified as a key factor in RA progression.

Summary: Chronic neutrophilic leukemia (CNL) and atypical chronic myeloid leukemia (aCML) are rare myeloid disorders with similar clinical presentations and outcomes. Mutational profiling revealed that both CNL and aCML have complex mutational profiles involving genes related to epigenetic regulation, splicing, and signaling pathways, with only EZH2 and TET2 differentially mutated between them. The study supports the idea that CNL and aCML are part of the same disease spectrum and highlights the importance of molecular-risk classification, particularly CSF3R mutations, in these diseases.

Summary: This study investigated the potential age, period, and birth cohort effects on the prevalence of suicide ideation in the European ageing population. The findings suggest that the relationship between age and suicide ideation is not linear in middle and older age, with the majority of variability attributed to individual-level factors. Birth cohort and period effects accounted for a small percentage of the total variability in suicide ideation, with Baby boomers born from the 1950s to mid-1960s reporting higher suicide ideation than their ancestors.
