Summary: TGFBR3L is a novel pituitary gland specific membrane protein, located in the membrane of gonadotroph cells in non-neoplastic anterior pituitary gland and in a subset of gonadotroph pituitary tumors. It is highly expressed in the pituitary and specifically detected in cells expressing FSH and LH, showing positive correlation with FSH and LH while inversely correlated with E-cadherin and estrogen receptor beta.

Summary: Pituitary adenomas (PAs) are mostly benign tumors, but up to 10% exhibit aggressive behavior. This article provides an overview of target structures in aggressive PAs and summarizes current clinical trials for treating PAs. Drug targets in PAs are based on general features of tumor cells and include immune checkpoints and signal pathways involved in tumor cell proliferation, migration, and angiogenesis. Dopamine agonists (DAs) are the first line treatment for prolactinomas and have potential to treat other subtypes of PAs. Targeting the invasive behavior of PAs may improve therapy.

Summary: Recent studies have focused on somatotroph pituitary adenomas in acromegaly patients showing a paradoxical GH response during OGTT, which has been linked to the ectopic expression of GIP receptor in adenoma cells. These adenomas are negative for activating GNAS gene mutations and have unique cytogenetic and DNA methylation profiles. Patients with paradoxical GH response pattern have higher IGF-1 levels and smaller adenomas, often densely granulated, and may respond better to somatostatin receptor ligands. Targeted therapy against GIP receptor on these adenomas could be a new treatment approach for these patients.
